黄芪、川芎及其配伍对脑缺血后凋亡相关基因表达影响的实验研究

目的 :探讨黄芪、川芎及其配伍对脑缺血后凋亡相关基因表达的影响。方法 :用结扎大鼠双侧颈内动脉的方法造脑缺血模型 ,采用免疫组化法分别检测bcl 2、P53 免疫阳性细胞。结果 :结扎大鼠双侧颈内动脉 1h后 ,脑皮质促凋亡质基因P53 表达明显增强 ,通过腹腔注射给药后 ,川芎对凋亡调控基因表达无影响 ,黄芪可抑制P53 表达 ,增强bcl 2表达 ,黄芪与川芎配伍用药在抑制P53表达方面明显优于单独给药。结论 :脑缺血后用黄芪配伍用芎可抑制缺血区P53 表达 ,增强bcl 2表达 ,在抑制P53 方面优于单独给药。

Experimental Researches of Astragalus Root Chuanxiong Rhizome and Them Used in Combination on Expression of Apoptosis Related Genes after Cerebral Ischemia Attack

Objective:To investigate the effects of Astragalus root,Chuanxiong rhizome and them used in combination on expression of apoptosis related genes after cerebral ischemia attack.Methods:make models on cerebral ischemia by ligating both of internal carotid artery,immunohistochemical method was used to observed bcl 2 and P 53 positive immunocytes.Results:It was found that the genes controlling apoptosis P 53 in cerebral cortex upregulated significantly after ligation of both.internal carotid artery for one hour,Under the adbominal administration,Chuanxiong rhizome didn't possess any effect on expression of genes controlling apoptosis.Astragalus root could inhibit expression of P 53 and enhance that of bcl 2,when Astragalus root and Chuan Xiong rhizome were used in combination the effects of inhibiting expression of P 53 were more obvious than those of single herb.Clusions:When Astragalus root and Chuan Xiong rhizome were used in combination.It could inhibit P 53 and enhance that of bcl 2,and the effects of inhibit expression of P 53 were more obvious than those of single herb.
