GDNF及其受体Ret在腺样囊性癌嗜神经侵袭和细胞增殖中作用的初步研究

摘    要:目的:探讨胶质细胞源性神经生长因子GDNF及其受体Ret的表达在涎腺腺样囊性癌(SACC)嗜神经侵袭和细胞增殖中的作用。方法:应用S-P法对42例SACC、5例腺泡细胞癌、5例正常涎腺组织中GDNF和Ret的表达进行检测。结果:GDNF在SACC肿瘤细胞的胞浆、肿瘤周围的神经纤维和正常腺体的导管中呈阳性表达,而在腺泡细胞癌和正常腺泡组织中均不表达。Ret在SACC肿瘤细胞的胞浆和正常的导管上皮细胞中呈阳性表达,在神经纤维中弱阳性或不表达,腺泡细胞癌中不表达。Ki-67在SACC细胞的胞核中呈阳性表达,靠近神经和远离神经的肿瘤细胞中阳性表达并无差异。有神经侵袭组Ki-67的阳性表达高于无神经侵袭组,但无统计学差异。GDNF与Ret表达间呈正相关(r=0.489,P〈0.05),但GDNF与Ki-67的表达间无明显相关性(r=0.278,P〉0.05)。结论:GDNF及其受体Ret在SACC中高表达,GDNF在周围神经组织内也高表达,提示SACC中存在GDNF的自分泌,GDNF和Ret可能在SACC嗜神经侵袭中发挥重要作用。此外,SACC细胞的增殖活性与神经侵袭的发生、GDNF的表达间关系可能不大。

Preliminary Study in Roles of GDNF and Ret in Perineural Invasion and Proliferation of Salivary Adenoid Cystic Carcinoma

Abstract:Objective:To investigate the roles of GDNF and Ret in perineural invasion and proliferation of salivary adenoid cystic carcinoma(SACC).Methods:Totally 42 SACC specimens,5 acinic cell carcinoma specimens,5 normal salivary tissues were included in the present study.S-P method was used to detect the expression of GDNF and Ret in SACC specimens,acinic cell carcinoma specimens,normal salivary tissues.Results:GDNF and its receptor Ret were highly expressed in SACC cells and normal ductual cells,and GDNF was also expressed in nerves adjacent SACC.The positive expression rates of GDNF,Ret in SACC were both 69.05%(29/42),while there was no expression in acinic cell carcinoma specimens and normal salivary acinus.Ki-67 was positively expressed in cytoplasm of SACC,with no statistical significance near and far from nerves,in PNI group and non-PNI group.Expression of Ret but not Ki-67 was relevant to GDNF(r=0.489,P0.05,r=0.278,P0.05,respectively).Conclusion:GDNF and Ret play important roles in the process of perineural invasion,while little function was found in the cell proliferation of SACC.Besides,there was autocrine of GDNF in SACC.
