不同pH值颠胃酸口服液的消化活力考察

目的：考察不同pH值颠胃酸口服液的消化活力,为其制备及储存提供参考。方法：制备颠胃酸口服液样品13组,pH值分别调整为1.0、1.5、2.0、2.5、3.0、3.5、4.0、4.5、5.0、5.5、6.0、6.5、7.0,常温下放置。以酪氨酸为对照品、牛血红蛋白为底物,于（37±0.5）℃水浴酶促反应,以275nm为检测波长,紫外分光光度法检测0、10、20、30、60d时的吸光度,计算胃蛋白酶消化活力。结果：0d时,不同pH值的颠胃酸口服液消化活力没有明显差异。60d时,pH为1.0～3.5的颠胃酸口服液平均消化活力为30.7U·mL-1,下降了24.6%,但均大于标示量（24U·mL-1）;而pH为4.0～7.0的颠胃酸口服液平均消化活力为21.6U·mL-1,下降了41.8%,均小于标示量。结论：pH值对颠胃酸口服液的消化活力有较大影响,常温下该溶液pH不超过3.5时可保存60d。

Investigation of Digestion Activity of Dianweisuan Oral Liquids with Different pH Values

OBJECTIVE：To investigate the activity of Dianweisuan oral liquids with different pH values and to provide reference for the preparation and storage of it.METHODS：13 groups of Dianweisuan oral liquids were prepared,and pH values were adjusted to 1.0,1.5,2.0,2.5,3.0,3.5,4.0,4.5,5.0,5.5,6.0,6.5 and 7.0 at room temperature.Use tyrosine as control,bovine hemoglobin as substrate by water bath-enzymic synthesis at（37±0.5）℃.The absorbance of oral liquids was determined on 0,10,20,30 and 60 d at the wavelength of 275 nm by UV spectrometry and the activity of pepsinum was calculated.RESULTS：On 0 day,there was no significant difference in the activity of Dianweisuan oral liquids with different pH values.On 60 d,the mean activity of oral liquids with pH value of 1.0～3.5 was 30.7 U·mL-1,decreasing by 24.6%,but they were more than labelled amount（24 U·mL-1）;the mean activity of oral liquids with pH value of 4.0～7.0 was 21.6 U·mL-1,decreasing by 41.8%,and they were lower than labelled amount.CONCLUSION：The results indicated that pH value has an apparent influence on the activity of Dianweisuan oral liquids.The oral liquids with pH value less than 3.5 can be preserved for 60 d at room temperature.
