不同pH值颠胃酸口服液的消化活力考察

摘    要:目的:考察不同pH值颠胃酸口服液的消化活力,为其制备及储存提供参考。方法:制备颠胃酸口服液样品13组,pH值分别调整为1.0、1.5、2.0、2.5、3.0、3.5、4.0、4.5、5.0、5.5、6.0、6.5、7.0,常温下放置。以酪氨酸为对照品、牛血红蛋白为底物,于(37±0.5)℃水浴酶促反应,以275nm为检测波长,紫外分光光度法检测0、10、20、30、60d时的吸光度,计算胃蛋白酶消化活力。结果:0d时,不同pH值的颠胃酸口服液消化活力没有明显差异。60d时,pH为1.0~3.5的颠胃酸口服液平均消化活力为30.7U·mL-1,下降了24.6%,但均大于标示量(24U·mL-1);而pH为4.0~7.0的颠胃酸口服液平均消化活力为21.6U·mL-1,下降了41.8%,均小于标示量。结论:pH值对颠胃酸口服液的消化活力有较大影响,常温下该溶液pH不超过3.5时可保存60d。

Abstract:OBJECTIVE:To investigate the activity of Dianweisuan oral liquids with different pH values and to provide reference for the preparation and storage of it.METHODS:13 groups of Dianweisuan oral liquids were prepared,and pH values were adjusted to 1.0,1.5,2.0,2.5,3.0,3.5,4.0,4.5,5.0,5.5,6.0,6.5 and 7.0 at room temperature.Use tyrosine as control,bovine hemoglobin as substrate by water bath-enzymic synthesis at(37±0.5)℃.The absorbance of oral liquids was determined on 0,10,20,30 and 60 d at the wavelength of 275 nm by UV spectrometry and the activity of pepsinum was calculated.RESULTS:On 0 day,there was no significant difference in the activity of Dianweisuan oral liquids with different pH values.On 60 d,the mean activity of oral liquids with pH value of 1.0~3.5 was 30.7 U·mL-1,decreasing by 24.6%,but they were more than labelled amount(24 U·mL-1);the mean activity of oral liquids with pH value of 4.0~7.0 was 21.6 U·mL-1,decreasing by 41.8%,and they were lower than labelled amount.CONCLUSION:The results indicated that pH value has an apparent influence on the activity of Dianweisuan oral liquids.The oral liquids with pH value less than 3.5 can be preserved for 60 d at room temperature.
