摘    要:目的:探讨大黄有效部位对慢性环孢素肾病(CCN)大鼠的保护机制.方法:雄性Wistar大鼠随机分为正常对照组;模型组;贝那普利阳性对照组;大黄有效部位干预1、2、3组.HE染色光镜观察肾组织病理变化情况,免疫组化法测定CD68、Ⅲ型和Ⅳ型胶原表达.结果:HE染色发现阳性对照组和大黄有效部位组其单核巨噬细胞浸润数比模型组明显减少;模型组肾组织的CD68、Ⅲ型胶原和Ⅳ型胶原表达高于阳性对照组和大黄有效部位干预组.结论:大黄有效部位可下调CCN大鼠CD68、Ⅲ型胶原、Ⅳ型胶原表达,表明其在延缓CCN肾间质纤维化进程中发挥着重要作用.

Pathogenesis Research of Rhubarb Effective Parts Prevention on Chronic Cyclosporine Nephropathy in Rats

Abstract:Objective: To investigate the promechanism of Rhubarb effective parts protection on chronic cyclosporine nephropathy (CCN). Methods : Male Wistar rats were randomly divided into normal control group, model group, benazepril positive control group, Rhubarb effective intervention part 1,2,3 groups. Renal tissue pathological changes were observated by HE dyed lens and CD68,11I type and 1V type collagen expression were examined by immunohistochemieal method. Results : HE stain found that the number of mononuclear macrophage infiltration in positive control group and rhubarb effective parts group were significantly reduced than that of the model group. CD68,III type and IV type collagen expression of the renal tissue in the model group were higher than positive control group and intervention group. Conclusion : Rhubarb effective parts can reduce the level of CD68, m type and 1V type collagen expression,which indicate that it has important role in CCN renal interstitial fibrosis progress.
