红景天玄参组合物对大鼠局灶性脑缺血再灌注损伤保护作用的研究

摘    要:目的探讨红景天与玄参组合物对局灶性脑缺血再灌注大鼠的保护作用。方法通过体外活性评价,筛选出红景天与玄参组合物最佳配比;采用颈内动脉线栓法建立局灶性脑缺血模型,观察模型大鼠的行为、脑梗死面积和脑含水量的变化;测定血清超氧化物歧化酶(SOD)活性和丙二醛(MDA)的含量。结果当红景天与玄参以1:1配比时,综合评分最好;且组合物中剂量组(0.39g/kg)、高剂量组(1.2g/kg)均能显著缩小局灶性脑缺血再灌注大鼠的脑梗死面积、降低梗死率,并能减少脑含水量(P〈0.05),同时使脑缺血72h大鼠的行为学症状得到显著改善(P〈0.05),明显提高SOD活性并降低MDA的含量(P〈0.05)。结论红景天与玄参组合物对局灶性脑缺血再灌注大鼠具有一定保护作用,且作用明显优于单用红景天或玄参。

Protective effect of the combination ofRhodiolae Crenulatae Radix et Rhizoma and Scrophulariae Radix on focal cerebral ischemia-reperfusion injury in rats

Abstract:Objective To investigate the protective effect of the combination of Rhodiolae Crenulatae Radix et Rhizoma and Scrophulariae Radix on focal cerebral ischemia-reperfusion injury of rats. Methods The in vitro activity evaluation was used to select the best ratio group of combination, and the internal carotid artery suture method was adopted to establish the model of focal cerebral ischemia model, and to observe the influence on the behavioral score, cerebral infarction area, and cerebral water content of model rats. Activity of superoxide dismutase (SOD) and content of malondialdebyde (MDA) in serum of rats were measured. Results The evaluation on the activity of the combination with the ratio in l:1 was the best, and the combination in the middle dose (0.39 g/kg), high dose (1.2 g/kg) groups could reduce the infarction area in the cerebral ischemia-reperfusion rats, decrease the infarction rate, reduce brain water content significantly (P〈0.05), at the same time, 72 h after modeling, The behavioral symptoms of the rats were improved significantly (P〈0.05). The combination could increase the activity of SOD and reduce the content of MDA (P〈0.05). Conclusion The combination ofRhodiolae Crenulatae Radix et Rhizoma and Scrophulariae Radix has a certain protective effect on the focal cerebral ischemia-reperfusion injury of rats, and the effect of the combination is superior to Rhodiolae Crenulatae Radix et Rhizoma alone or Scrophulariae Radix.
