冷刺激豚鼠食道诱导气道血浆渗出及非肾上腺素能非碱胆能神经的作用

摘    要:目的 观察非肾七腺素能非碱胆能神经在冷刺激豚鼠食道诱导气道血浆渗出中的作用.方法 豚鼠麻醉后,食管上下端插管,食管灌注4℃生理盐水.根据分组需要静脉注射阿托品及心得安阻断胆碱能及肾上腺素能通路,观察气管、主支气管、细支气管中微血管血浆渗出情况,以及SP受体拮抗剂FK888干预或预先切断迷走神经对微血管血浆渗出的影响.气道血浆渗出采用伊文思蓝法.结果 冷刺激豚鼠食道显著增加气管、主支气管、细支气管的血浆渗出(P<0.01).SP受体拮抗剂FK888显著抑制冷刺激诱导的气管和主支气管血浆渗出(P<0.05).双侧迷走神经切除后,冷刺激豚鼠食道引起的气管、主支气管血浆渗出被显著抑制(P<0.05).结论 冷刺激食道可以诱导气道血浆渗出,其机制可能与非肾上腺素能非胆碱能通路有关.

Abstract:Objective To observe the role of nonadrenergic noncholinergic nerve in airway microvascular leakage induced by cold sodium chloride intra-oesophageal instillation in guinea pigs. Methods Guinea pigs were anaesthetised with pentobarbital sodium, a catheter was placed in the midoesophagus and distal esophagus, infusion of 4℃ NS into the esophagus. Introvenous injection of atropine and propanolol to block the cholinergic and adrenergic nerves according to the groups. The microvascular leakage of trachea,main bronchus,bronchiole was examined in the presence and absence of bilateral vagus nerve or NK-1-receptor antagonist FK888. The airway plasma leakage was evaluated by measuring extravasated Evans blue dye. Results Infusion of 4℃ NS into the esophagus significantly increased the tracheal,mainstem bronchial and bronchiolar Evans blue dye concentration ( P<0.01). The tracheal and mainstem bronchial plasma extravasation induced by intra-oesophageal 4℃ NS instillation were significantly inhibited by bilateral vagotomy and FK888 ( P<0.05). Conclusions Cold stimulation in oesophagus can induce airway microvascular leakage, which may be mediated by nonadrenergic noncholinergic nerve.
