苯肾上腺素延长糖尿病大鼠心脏QT间期的机理研究

摘    要:目的:分析苯肾上腺素显著延长糖尿病大鼠心脏QT间期的钾电流机理。方法:采用膜片钳全细胞式的记录方法,观察苯肾上腺素对正常及糖尿病大鼠左心室肌细胞的瞬时外向钾电流(Ito)及稳定状态的钾电流(Iss)的差异作用。结果:与对正常动物的作用相比,苯肾上腺素降低正常的和糖尿病大鼠的瞬时外向钾电流(Ito),和稳定状态的钾电流(Iss)。但与正常大鼠相比,降低糖尿病大鼠稳定状态的钾电流(Iss)更明显。结论:与正常动物相比,苯肾上腺素明显延长糖尿病大鼠离体心脏QT间期及动作电位复极90%水平时程(APD90)是由于其更显著降低左心室肌细胞稳定状态的钾电流(Iss)。

Abstract:Aims:To analyze the ionic mechanisms underlying prolongation of rate-corrected QT intervals (QTc) and action potential duration at 90% level (AP90) by phenylephrine in diabetic rats.Methods: To observe the differential effects of phenylephrine on the transient outward K current (Ito) and the steady state K current (Iss) in normal and diabetic rats using patch clamp whole-cell configuration.Results:Phenylephrine significantly reduced Ito and Iss in both normal and diabetic rats but the Iss reduction rate was greater in diabetic rats than in normal rats.Conclusion: The greater prolongation rates of QTc and AP90 induced by phenylephrine in diabetic rats was mediated by greater reduction in the rate of Iss in diabetic rats
