注射用益气复脉(冻干)后给药对脂多糖引起的大鼠肠系膜微循环障碍的改善作用

摘    要:观察注射用益气复脉(冻干)后给药对脂多糖(lipopolysacchride,LPS)引起的大鼠肠系膜微循环障碍的改善作用。方法:用微循环观察系统记录Wistar雄性大鼠肠系膜微循环动态。静脉滴注LPS 20min后,静推注射用益气复脉(冻干)(小剂量30mg/kg、中剂量80mg/kg、大剂量160mg/kg),90min后计数肥大细胞脱颗粒率。结果:注射用益气复脉(冻干)的小剂量后投入,在投入60min时抑制LPS引起的细静脉过氧化物的产生。中剂量和大剂量的后投入抑制了细静脉血管壁白细胞的粘附和过氧化物产生。三个剂量的后投入均能显著抑制肥大细胞脱颗粒。结论:注射用益气复脉(冻干)后投入通过抑制白细胞与细静脉的粘附,抑制过氧化物产生和抑制肥大细胞脱颗粒改善了LPS引起的大鼠肠系膜微循环障碍。

Abstract:This study is designed to observe the attenuation effects of post-treatment with Zhusheyong Yiqifumai on lipopolysacchride (LPS)-induced rat mesenteric microcirculation disturbance.
In the study, the hemodynamics of mesenteric microcirculation of male Wistar rats was observed under an inverted biological microscope. 20 min after LPS infusion via vein, Zhusheyong Yiqifumai was administrated in one shot (small dosage at 30 mg/kg, medium dosage at 80 mg/kg, and high dosage at 160 mg/kg). 90 min after observation, the mast cell degranulation in the mesenteric interstitium was counted.
It is found that the post-treatment in small dose of Zhusheyong Yiqifumai noticeably inhibited the production of LPS-induced oxygen free radicals from venular wall 60 min after the injection. The post-treatment with a medium or high dosage of Zhusheyong Yiqifumai significantly inhibited leukocyte adhesion and output of oxygen free radicals in mesenteric venule. All dosages have been proved with an inhibitory effect on the LPS-induced mast cell degranulation. The study demonstrates that the post-treatment with Zhusheyong Yiqifumai has an ameliorating effect on LPS-induced microcirculation disturbance in rat mesentery. The effect is probably associated with the inhibitory effects on leukocyte adhesion to venular wall, oxygen free radicals production, and mast cell degranulation.
