大鼠骨髓间充质干细胞移植后在溃疡性结肠炎模型肠道的定位

摘    要:目的:探讨同种异体大鼠骨髓间充质干细胞(MSCs)移植于大鼠溃疡性结肠炎(UC)模型后其在结肠的定位情况.方法:将用贴壁法体外连续传代培养的雄性SD大鼠的MSCs经尾静脉注射于雌性大鼠UC模型体内,并以UC模型组及酒精灌肠为对照组.分别于移植后第1天、第7天、第21天后取远端结肠组织,经分子生物学方法检测Y染色体的性别决定区段(sry)作为标志,以确定MSCs的肠道定位情况.结果:培养的MSCs集落生长迅速,均一性好.TNBS引起肠壁明显增厚,炎症和溃疡持续时间较长,髓过氧化物酶活性值显著性升高,不同时期均与对照组有明显差异(P<0.01),黏膜及黏膜下层有大量中性粒细胞、淋巴细胞、巨噬细胞及纤维细胞浸润.移植组的sry检测显示:第1天,第7天和第21天阳性率分别为0,16.7%(1/6)和50.0%(3/6),而对照组均阴性.结论:移植后的MSCs能在UC模型的肠道中定位.

Abstract:Objective:To investigate the localization of bone marrow mesenchymal stem cells(MSCs) in the colon of allografted rat models of ulcerative colitis.Methods: The MSCs from SD male rats were purified by culture-expansion in vitro and then were transplanted into SD female rat models of ulcerative colitis(TNBS-produced) through caudal vein.Meanwhile,UC model group and alcohol enema group were used as controls.The sex-determining gene(sry) located on Y chromosome was examined by PCR in colon of female rats on days 1,7 and 21 after transplantation,respectively,to localize the colon MSCs.Results: Rapid growth with good homogeneity in cultured MSCs was observed.TNBS caused significant mural thickening of colon,prolonged inflammation and ulcers,and markedly elevated MPO activity,with statistical differences between varied time spots and between groups.The expression rate of the sry gene was 16.7%(1/6) on day 7 and 50.0%(3/6) on day 21 in the allografted models.But all the control groups are sry-negative.Conclusion: MSCs can be localized to the colon of ulcerative colitis rat models.
