银杏叶提取物对新生大鼠缺氧缺血性脑损伤后Survivin表达的影响

摘    要:目的观察银杏叶提取物(extract of ginkgo biloba,EGB)对新生大鼠缺氧缺血性脑损伤(hypoxic—ischemic braindamage,HIBD)后缺血侧脑组织Survivin表达的影响,探讨其神经保护机制。方法健康7日龄Sprague—Dawley新生大鼠72只随机分为假手术组(n=24)、对照组(n=24)及EGB治疗组(n=24)。采用经典的PAce法制成HIBD模型,应用TUNEL法检测细胞凋亡情况。并采用免疫组织化学二步法检测Survivin表达的变化。结果与假手术组相比,对照组伤后1、3、7、14d的细胞凋亡数及Survivin表达水平均明显升高(P〈0.01);与对照组相比,EGB治疗组伤后7、14d的细胞凋亡数明显降低(P〈0.01),且在伤后1、3d的Survivin表达明显升高(P〈0.05)。结论EGB可促进抗凋亡因子Survivin的表达而减少HIBD后的细胞凋亡,发挥神经保护作用。

Abstract:Objective To explore the effect of EGB on the expression of survivin after hypoxic-ischemic brain damage (HIBD) in neonatal rats and the mechanisms of neuroprotection by EGB. Methods Seventy-two seven-day-old Sprague-Dawley rats were randomly divided into sham operation group (n=24), HIBD group (n=24) and EGB group (n=24). HIBD was induced by the ligation of left common carotid artery, followed by a 2-hr exposure to 8% 02. The ischemic cerebral hemisphere were obtained 1, 3, 7 and 14 days after HIBD for determining apoptosis of cells by TUNEL and the expression of survivin protein by immunochemical method. Results Compared to the sham operation group, the number of apoptotic cells and survivin immunopositive cells increased significantly in the ischemic cerebral hemisphere 1, 3, 7 and 14 days after HIBD (P〈0.01). EGB increased significantly the expression of survivin (P 〈0.01), and decreased significantly the apoptotic cells compared to HIBD group (P 〈0.05). Conclusion The expression of survivin increased by EGB might be one of the mechanisms in inhibiting neuronal apoptosis in the ischemic cerebral hemisphere.
