HLA—A2表达沉默的MSCs对兔桡骨缺损修复的影响

摘    要:目的探讨沉默人白细胞抗原-A2(HLA-A2)基因后的骨髓间充质干细胞(MSCs)对兔桡骨缺损修复的影响。方法将HLA-A2沉默后的MSCs经体外诱导成骨后,与羟基磷灰石(HA)复合培养,形成MSCs/HA复合体,并植入骨缺损中。将日本大耳白兔24只分为2组:HLA-A2沉默后的MSCs/HA复合体修复桡骨缺损为实验组(n=12),自体MSCs/HA复合体修复桡骨缺损为对照组(n=12)。于术后2、4、8周比较两组饮食与伤口的愈合情况、X线检测、HE染色结果,综合评价HLA-A2沉默后的MSCs/HA复合体对桡骨缺损修复的影响。结果实验组和对照组的伤口无明显渗出、红肿,愈合好;X线结果显示,2、4周骨缺损处有纤维骨痂形成,8周骨缺损处密度增高,骨痂形成;两组细胞钙结节数量、X线评分、组织学评分比较差异无统计学意义(P〉0.05)。结论 HLA-A2沉默后的MSCs免疫原性低,不影响其成骨能力及骨缺损的愈合,为同种异体骨缺损修复提供了新的依据。

Abstract:Objective To explore after the HLA-A2 gene silencing of the effect of bone marrow mesenchymal stem cells (MSCs) on the repair of rabbit radial bone defect. Methods After the silence HLA-A2 MSCs by in vitro induction of osteogenesis, and togther Hydroxyapatite composite culture to form the MSCs/HA complex, and implant of bone defect. The rabbits were divided into 2 groups:silence after HLA-A2 MSCs/HA complex repair bone defect as experimental group(n=12), autologous MSCs/HA complex to repair bone defect as control group(n=12). Respectively after 2, 4, 8 weeks were compared: diet and wound healing, X-ray detection, HE staining results, comprehensive evaluation of silence after the HLA-A2 MSCs/HA complex on bone defect repair effect. Results Both of experimental group and control group had no obvious exudation of wound, swelling, obvious healing; X-ray showed: 2, 4 weeks bone defect with fibrous callus formation, 8 weeks bone defect density increased, callus formation, without significant difference between the two groups (P〉0.05). Conclusions HLA-A2 expression silencing of the MSCs has low immunogenicity,it does not influence the osteogenic ability and bone defect healing.This is provides a new evidence for allogeneic bone defect repair.
