大鼠肝抑素提取物生物效应的体外研究

应用酒精深山析和超速离心等方法人正常成年大鼠中提取肝抑素粗制品。离体实验检测提取物对再生肝肝细胞，、肝癌细胞、胃癌细胞的生物效应，结果是：（１）肝抑素提取物对再生肝肝细胞增殖具有很强抑制作用。随剂量的增加，细胞增殖抑制率增大，其最大抑制率为７１．４％（２）对肝癌细胞同样具有抑制作用，其最大抑制率为４３％；对人胃癌细胞无抑制作用。表明提取物中含有肝抑素，经对培养再生肝肝细胞、人肝癌细胞的增殖具有抑制

BIOLOGICAL EFFECTS OF CRUDE HEPATIC CHALONE EXTRACTED FROM RAT LIVER IN VITRO

With alcohol precipitation and ultracentrifugation,we extracted crude hepatic chalone(HC) from normal rat liver,and assayed its biological activity to the regenerating liver cells,the human hepatoma cells and the gastric cancer cells in vitro. The results were as follows:(1)The HC appeared to have strong inhibition on regenerating hepatocytes.The higher the doses were,the greater the inhibitory rate was.The maximal inhibition was about 71.4%.(2)The HC could also inhibit the growth of human hepatoma cells,but the cells were less sensitive than that of regenerating cells,the maximal inhibition was 43%;and it appeared to have no inhibitory effect on human gastric cancer cells.These rasults indicate that the crude extract contains hepatic chaione.It can both inhibit proliferation of regenerating hepatocytes and hepatoma cells,but the inhibition on regenerating hepatocytes is greater than that on hepatoma cells;and it also appeared that the HC have the tissue specificity to the hepatoma cells.
