肝癌组织中PTEN的表达及其与p53、细胞增殖和凋亡的关系

摘    要:目的:研究肝细胞肝癌(HCC)组织中PTEN的表达与细胞增殖及凋亡的关系及其与p53的相关性,明确PTEN是否参与HCC的发生、发展. 方法:利用免疫组织化学技术检测47例经术后病理学证实的HCC组织、42例癌旁肝组织及10例健康肝组织中PTEN,p53,增殖性核抗原(PCNA)的表达;用TUNEL法检测HCC中细胞凋亡水平. 结果:10例正常肝组织、42例癌旁肝组织中PTEN全部阳性表达(100%);53%(25/47)的HCC组织PTEN蛋白表达缺失. PTEN的表达与肝癌细胞的增殖指数和凋亡指数无相关性,与肝癌的浸润转移具有相关性,与p53的表达有相关性. 结论:肿瘤抑制基因PTEN和p53有相互作用. PTEN蛋白表达缺失可能在HCC的进展过程中发挥主要作用.

Abstract:AIM: To investigate the relationship of the expression level of PTEN protein with p53 cell apoptosis and proliferation in hepatocellular carcinoma (HCC) tissue so as to explore its roles in the development of HCC. METHODS: Immunohistochemistry was used to assess the protein expression of PTEN, PCNA and p53 in HCC tissues from 47 cases pathologically confirmed after operation, in adjacent normal liver tissues from 42 cases and in 10 normal liver tissues. TUNEL assay was performed to detect apoptosis in HCC tissues. RESULTS: PTEN protein expression was found in the 10 normal tissues and the adjacent normal liver tissues from the 42 cases while no PTEN protein expression was detected in HCC tissues from 25 cases pathologically confirmed after operation. The expression of PTEN was correlated to the metastasis of HCC and p53 expression. There was no correlation between PTEN protein with apoptosis index and proliferation index. CONCLUSION: PTEN, which acts as a tumor suppressor gene, interacts with p53. The lack of PTEN protein expression in HCC may suggest an important role of PTEN in the progression of HCC.
