摘    要:目的 探讨小鼠胶质瘤细胞系GL261主要高表达的Toll样受体(TLRs)家族成员及其在GL261细胞迁移中的作用.方法 运用实时荧光定量PCR检测TLRs在GL261细胞的表达情况;GL261细胞经TLR2配体Pam3CSK4刺激后,应用Transwell迁移实验检测其对迁移能力的影响;慢病毒稳定干扰TLR2表达后,观察Pam3CSK4刺激对GL261细胞迁移能力的改变.结果 GL261细胞主要高表达TLRs家族中的TLR2;Pam3CSK4激活TLR2后显著增强GL261细胞迁移能力;沉默TLR2基因表达后,Pam3CSK4促进GL261细胞迁移能力的作用消失.结论 小鼠GL261胶质瘤细

Abstract:Objective To explore the expression of Toll-like receptor family(TLRs)in mouse glioma cell line GL261 and effects of TLR2 activation on migration capacity of GL261 cells. Methods The expression of TLRs in GL261 was detected by qRT-PCR. Transwell migration assays were performed to determine the migration capacity of GL261 cells stimulated with Pam3CSK4, a TLR2 ligand. The change of migration capacity after silencing TLR2 in GL261 was examined by transwell assays. Results TLR2 of the TLRs family was highly expressed in GL261 cells. Pam3CSK4 stimulation enhanced migration capacity of GL261 cells was induced. Conclusion Triggering of TLR2 expressed by GL261 cell line enhanced its migration capacity, and the effect was reduced with down-regulation of TLR2.
