芍药苷对黑素细胞生物活性的影响

摘    要:目的:检测芍药苷对体外培养正常人表皮黑素细胞增殖活性、酪氨酸酶活性及黑素合成的影响。方法:建立正常人表皮黑素细胞培养体系,加入不同浓度(5~640μg/mL)芍药苷并作用一定时间后,采用MTT法、体外氧化DOPA反应法、NaOH法等分别测定黑素细胞增殖活性、酪氨酸酶活性及黑素含量的变化。结果:40~160μg/mL芍药苷呈剂量依赖性抑制黑素细胞增殖,与对照组比较差异有统计学意义(P0.05);5-20μg/mL芍药苷对酪氨酸酶活性呈剂量依赖性促进作用,与对照组比较差异有统计学意义(P0.05);10-20μg/mL芍药苷对黑素细胞黑素合成起促进作用,与对照组比较差异有统计学意义(P0.05)。结论:芍药苷在较高浓度范围内(40~160μg/mL)对体外培养正常人表皮黑素细胞增殖有剂量依赖性抑制作用;在较低浓度范围内(10-20μg/mL)对其黑素合成有促进作用。

Effect of paeoniflorin on biological activity in normal human malanocytes in vitro

Abstract:Objective:To investigate the effect of paeoniflorin on cell proliferation,tyrosinase activity and melanin synthesis in cultured normal human melanocutes. Methods:After paeoniforin (5-640 μg/mL) was added into cultured normal human malanocytes,the effect of paeoniflorin on the proferation of melanocytes,ty-rosinase activity and synthesis of melanin in melanocytes were measured by MTT method,dopamine oxidation method and NaOH method,respectively. Results:40-160 μg/mL paeoniflorin inhibited the growth of mela-nocytes in a concentration-dependent manner ( P〈0.05) . When the concentrations of 5-20μg/mL paeonifor-in were added,the tyrosinase activity was significantly increased in a concentration-dependent manner ( P〈0.05). When the concentrations of 10 -20μg/mL paeoniforin were added,the melanin synthesis were signifi-cantly increased ( P〈0.05) . The differences in the results mentioned above among the test groups and control group were significant (all Ps〈0.05).Conclusion: Paeoniflorin in concentrations of 40-160 μg/mL can in-hibit the proliferation in cultured normal human malanocytes in a concentration-dependent manner. When the concentration of 10-20 μg/mL is used the melanin synthesis is promoted.
