补阳还五汤对大鼠局灶性脑缺血后海马齿状回神经干细胞增殖和存活的影响

摘    要:[目的]观察补阳还五汤对大鼠局灶性脑缺血后海马齿状回神经干细胞增殖和存活的影响。[方法]采用线栓法诱导大鼠大脑中动脉阻塞模型,缺血90min后再灌,缺血后24h开始灌胃补阳还五汤(13g.kg-1),1次/d,缺血后第5~7天腹腔注射Br-dU(50mg.kg-1,2次/d,间隔8h),分别在缺血后第8和35天处死,免疫组化检测海马齿状回BrdU阳性细胞。[结果]与模型组比较,缺血后第8、35天,补阳还五汤组BrdU阳性细胞数显著增加(P〈0.01)。[结论]补阳还五汤能促进脑缺血大鼠海马齿状回神经干细胞增殖和存活。

Abstract:[Objective] To explore the effect of Buyanghuanwu decoction on proliferation and survival of neural stem cells in the dentate gyrus after focal cerebral ischemia in rats.[Methods] Focal cerebral ischemia was induced by occlusion of the right middle cerebral artery(MCA) for 90 min using the intraluminal filament model.Buyanghuanwu decoction treatment(13 g·kg-1,orally) was started on day 1 after ischemia once a day.BrdU was injected(i.p.50 mg·kg-1,twice a day,interval 8 h) on days 5-7 after ischemia.BrdU positive cells were examined by immunohistochemistry on days 8,35 after ischemia,respectively.[Results] The number of BrdU-positive cells in the dentate gyrus was significantly enhanced following Buyanghuanwu decoction treatment 8 days after ischemia.In addition,the number of surviving BrdU-positive cells also increased markedly in the Buyanghuanwu group 35 days after ischemia.[Conclusion] Buyanghuanwu decoction treatment after ischemia improves proliferation and survival of neural stem cells in dentate gyrus following focal cerebral ischemia,which may be the mechanism for the enhancement of functional recovery.
