清瘟败毒饮对急性肺损伤大鼠促炎症和抗炎症因子水平的影响

摘    要:目的]观察清瘟败毒饮对脂多糖(LPS)诱导急性肺损伤(acute 1ung injury,ALI)大鼠血清炎症因子IL-1β、IL-13水平的影响.方法]144只SD大鼠随机分为正常对照组,模型组,泼尼松组和清瘟败毒饮大、中、小剂量组(n=24只).造模后连续用药3d,2次/d.于末次给药后24h、48h每组各处死6只大鼠,72h处死余下所有大鼠,ELISA法检测血清炎症因子IL-lβ和IL-13的含量;HE染色观察肺组织病理变化.结果]模型组各时相血清中IL-1β和IL-13水平明显高于空白对照组(P<0.01),但IL-1β增加的速度明显高于IL-13;与同时相模型组相比,清瘟败毒饮各剂量治疗组(除24h小剂量组外)大鼠血中IL-1β含量均下降,IL-13含量均升高(P<0.05或P<0.01);LPS致ALI大鼠肺组织病理检查结果显示肺泡结构破坏或实变,肺泡隔增厚显著,毛细血管充血明显,肺泡腔内、细动脉周围和细支气管壁可见成堆炎症细胞浸润,而清瘟败毒饮干预组肺组织病理均有较大程度的改善.结论]清瘟败毒饮可通过有效调节脂多糖致急性肺损伤大鼠血中炎症细胞因子IL-lβ和抗炎症细胞因子IL-13表达水平,促使炎症和抗炎症细胞因子趋于动态平衡,从而减轻肺部炎症细胞浸润,起到修复保护损伤肺组织的作用.

Abstract:Objective]To observe Qingwen Baidu decoction,the traditional Chinese medicine,on LPS-induced acute lung injury(ALI) lung tissue pathology and the expression of IL-1β,IL-13.Methods]144 healthy male SD rats were randomly divided into 6 groups(n=24):normal control group,model group,the prednisone group and the Qingwen Baidu decoction high,middle and low dose groups.The LPS solution(2mg/kg) was instilled into the tracheal to prepare rat ALI model,once a day for two days.Each treatment group completed the second infusion 1 hour after LPS solution,the drug with saline diluted to the appropriate 4ml to intragastric administration twice a day.After each group were fed 24h,48h,6 rats were put to death,all remaining rats were sacrificed at 72h,and to collect the required samples.We observed the pathological changes of lung tissue under light microscope,and detected the IL-1β and IL-13 expression in lung tissue by ElISA.Results]At each time of serum IL-1β and IL-13 levels in rats of model group were significantly higher than the control group(P<0.01),but the rate of increase in IL-1β was significantly higher than IL-13;the content of IL-1β in rats serum decreased(except low dose group of 24h),and the IL-13 in blood serum rose in all doses of Qingwen Baidu decoction treatment groups.Compared with the model group,phase has significant or very significant difference(P<0.05,P<0.01);The pathology of rats lung tissue with LPS induced,ALI showed pulmonary alveolus structure of the blank group was complete and without edema,and the pulmonary alveolus septum wasn’t incrassated,occasionally there’s some inflammatory cellular infiltration.While pulmonary alveolus structure of the model group was wrecked or with consolidation,and the pulmonary alveolus septum was incrassated obviously,the blood capillary hyperemia was remarkable,and piles of inflammatory cell could be seen in the alveolar cavity and around the thin artery and on the bronchiole wall.The intervention group Qingwen Baidu Decoction lung pathology than the model group had a greater degree of improvement.Conclusion]Qingwen Baidu Decoction can effectively regulate cytokine IL-1β and anti-inflammatory cytokine IL-13 expression levels in serum of LPS-induced acute lung injury rats and promote inflammation and anti-inflammatory cytokines homeostasis tends to reduce the pulmonary inflammatory cells,play a protective injury repair role of lung tissue.
