刺五加皂苷脂质体的制备及对肝癌移植瘤的抑制作用

摘    要:目的:制备刺五加皂苷脂质体并研究其对肝癌移植瘤的抑制作用。方法:采用薄膜分散-超声法制备刺五加皂苷脂质体并检测其理化性质,用流式细胞仪检测此载药脂质体对HepG2肝癌细胞周期和凋亡的影响,并观察刺五加皂苷脂质体荷瘤小鼠的抑瘤作用。结果:所制备的脂质体封率为63.43%,刺五加皂苷脂质体组与游离刺五加皂苷组均使肝癌细胞G0/G1期和S期比例明显下降,并观察到细胞凋亡,高剂量刺五加皂苷脂质体组荷瘤小鼠的抑瘤率(62.21%)显著高于低剂量组(45.23%)和游离刺五加皂苷组(46.16%,P均<0.05),而低剂量刺五加皂苷脂质体组与游离刺五加皂苷组无明显差异(P>0.05)。结论:刺五加皂苷脂质体能抑制肝癌细胞Hep G2增殖和诱导细胞凋亡,对小鼠肝癌移植瘤的生长有明显抑制作用。

Abstract:Objective:To prepare Acanthophopanaxenticosus(ASS) liposomes and investigate its therapeutic effect on implanted hepatocellular carcinoma.Methods:The Acanthophopanaxenticosus liposomes were prepared by the thin-film hydration and ultrasonic technique.The flow cytometry were applied to study cell cycle and apoptosis of the hepatocellular cancer cell line Hep G_(2)and the tumor growth inhibition rate in micewere calculated.Results:The ASS liposomes encapsulation percentage is 63.43%.The cell number rate in G_(0)/ G_(1)phase and S phase were reduced significantly.The inhibitory effect of high-dose ASS liposomes group(62.21%) was stronger than that of low-dose ASS liposomes group(45.23%)or single ASS group(46.16%,P<0.05);Compared with the single ASS group,the inhibition rate in the low-dose ASS liposomes group was no significantly difference.Conclusion:The ASS liposomes can inhibit significantly proliferation of hepatocellular on tumor growthof cancer cell Hep G_(2) and induce apoptosis,can also inhibit tumor growth in mice.
