糖尿病性肢端坏疽大鼠模型的建立及实验研究

摘    要:目的:建立一种具有实验价值的糖尿病性肢端坏疽的大鼠模型并进行实验研究。方法:给雄性Wistar大鼠腹腔注射中等剂量的链脲佐菌素后,适当调降室温,以建立动物模型,同时对治疗组用自拟气阴两虚中药复方加以治疗:连续观察测量实验期间各组的饮水量及体表变化,测定各组在造模前后的空腹血糖、三酰甘油、胰岛素和体重。结果:糖尿病组较正常对照组有明显的肢端坏疽出现、体重明显下降,空腹血糖、三酰甘油升高,而空腹胰岛素水平明显下降;同时糖尿病治疗组与糖尿病对照组相比:肢端坏疽明显减轻,体重明显升高,空腹血糖、三酰甘油明显下降,而空腹胰岛素水平明显上升。结论:血清胰岛素水平升高,血糖、血脂水平下降,可在一定程度上防治糖尿病足的发生、发展;同时,也证明此模型对药物反应敏感,可作为研究糖尿病足发病机理及药物治疗效果评价的动物模型。

Abstract:Objective To establish a valuable rat model with diabetic foot and study it. Methods In order to establish an animal model with diabetic foot diabetic foot,male Wistar rats were injected with moderate dose of Steptozotocin ,and the room temperature was down regulated appropriately for 7 days. At the same time,the treatment group was treated with Yiqi Yangyin Recipe. During the experimental period, the changes of water drinking amount, syndrome of body surface were observed and measured in both groups. Fasting plasma glucose, triglyceride,insulin levels and body weight were also measured before and after the animal models were established in two groups. Results Compared to normal group,diabetic foot group had significant syndrome of diabetic foot,greatly decreased body weight,increased fasting plasma glucose and triglyceride,but the levels of insulin were significantly lowered. Meanwhile,compared to diabetic foot control group, syndrome of diabetic foot signficaantly mitigated,body weight greatly increased,fasting plasma glucose and triglyceride decreased,but the levels of insulin significantly increased in diabetic foot treatment group. Conclusion To some extent, increase of insulin level and decrease of fasting plasma glucose and triglyceride may prevent diabetic foot from beginning and developing, which suggests that diabetic foot rat model established by our method be sensitive to medicine,and it could be used to study the mechanism of diabetic foot and to evaluate the therapeutic effect of some medicine.
