摘    要:目的:探讨刺蒺藜总皂苷对急性药物性肝损伤的防治作用及其机理。方法:60只小鼠随机分为模型组,刺蒺藜总皂苷低、中、高剂量组,甘利欣对照纽和空白组,分别以相应剂量药物灌胃7d,然后以雷公藤多苷灌胃1次造模,观察小鼠血清AIJT、AST水平变化及肝组织Caspase-3表达情况。结果:刺蒺藜总皂苷可有效降低小鼠血清ATJT、AST水平,与模型组比较,有显著统计学意义(P〈0.05),并使肝组织Caspase-3表达降低,尤以高、中剂量组效果最佳。结论:刺蒺藜总皂苷对雷公藤多苷诱导的急性肝损伤具有较好保护作用,其机理与降低Caspase-3表达、抑制肝细胞凋亡有关。

Abstract:Objective :To explore the preventive and therapeutic effect and mechanisms of total saponins from tribulus terrestris(TSTT) on acute drug-induced liver injury. Methods :60 mice were randomly divided into model group,low-dosage, medium-dosage and high -dosage group of TSTT, Ganlixin group and control group. Each group was respectively perfused with corresponding drug for 7 days and then tripterygium glycosides were perfused once to make models. The levels of ALT and AST in the serum were respectively measured and Caspase-3 expression of liver tissue was observed. Results :The levels of ALT and AST in the serum of each TSTI' group reduced obvi- ously. Compared with model group ,the differences were statistically significant( P 〈 0.05 ). Meanwhile ,TSTI' could reduce Caspase-3 expression of liver tissue. The effects of high-dosage group and medium-dosage group were especially conspicuous. Conelusion:TSTr has better protective effect on acute liver injury induced by tripterygium glycosides, the mechanism of which is mainly related with reducing Caspase-3 expression and inhibiting apoptosis of the liver cells.
