高乌甲素凝胶透皮吸收实验皮肤的选择研究

摘    要:目的:选择适当的动物皮肤,建立适合于高乌甲素凝胶的透皮吸收研究方法。方法:高数液相色谱HPLC法测定高乌甲素的含量;采用改良Franz扩散池进行透皮吸收实验,以累积渗透量对取样时间进行回归,计算透皮速率常数;从裸鼠、小鼠、大鼠、家兔、乳猪皮肤及人皮中筛选实验皮肤。结果:裸鼠皮肤的透皮吸收速率常数与人皮较为接近,且重复性较好。结论:通过对高乌甲素透皮吸收实验皮肤的选择,为研究其TDD制剂提供了方法学依据。

Abstract:Objective:To select proper animal skin and establish effective research methods of percutaneous endosmosis for lappaconitine gel.Methods:The concentration of lappaconitine was determined by HPLC;Improved Franz-type diffusion cell was used for the percutaneous absorption study.Percutaneous endosmic speed constant and the accumulative amount of percutaneous endosmic drug were used for the estimation of animal skin selection from hairless mice,rats,rabbits,pig ears and human.Results:The percutaneous endosmic speed constant of hairless mouse skin was much similar to that of human,and the obtained results were reliable and repeatable.Conclusion:The selection of the animal skin for the percutaneous endosmic of lappaconitine gel provided methodological reference for TDD preparations.
