p73在大肠癌组织中的表达及其意义

摘    要: 目的 探讨 p73 表达与大肠癌生物学行为、预后的关系。 方法 研究对象为 60 例术前未经放疗、化疗的大肠癌患者的癌组织标本及其中 23 例患者的癌旁组织标本。60例患者中高分化腺癌 21 例,中分化 22 例,低分化 17 例;侵及浆膜者 36 例;有淋巴结转移者 21 例。应用原位杂交技术检测 p73 mRNA 的表达,应用免疫组织化学法检测 p73蛋白的表达,利用 Smartscape 图像分析系统进行定量分析,结果以平均灰度值表示,平均灰度值低示表达量高。 结果 p73 mRNA及p73蛋白平均灰度值在大肠癌组织分别为0.549 ± 0.080 及 0.481 ± 0.091,在癌旁组织中分别为 0.703 ± 0.081 和 0.701 ±0.068,二者在癌组织中的表达量均高于癌旁组织(P < 0.05)。将患者按肿瘤分化程度、是否侵及浆膜及有无淋巴结转移分组,p73 mRNA及p73 蛋白平均灰度值在低中分化组分别为 0.463 ± 0.082和0.472 ± 0.099,高分化组分别为 0.549 ± 0.088 及0.535 ± 0.0584;在侵及浆膜组分别为 0.517 ± 0.106 和0.528 ± 0.100,未侵及浆膜组分别为 0.602 ± 0.079 和0.560 ± 0.058;在淋巴结转移组分别为 0.513 ± 0.083 及 0.462 ± 0.105,无淋巴结转移组分别为0.607 ± 0.083 及 0.595 ± 0.080。各组间 p73 mRNA及 p73 蛋白平均灰度值的差异均有统计学意义(P < 0.05)。 结论 p73 在大肠癌组织中的表达增强,并与肿瘤的生物学行为相关,可作为判断大肠癌进展和预后的指标。

Expression and significance of p73 mRNA and protein in colorectal carcinoma

Abstract:Objective To investigate the association between the expression of p73 and the biological behavior and prognosis of colorectal carcinoma. Methods A total of 60 samples of carcinoma tissues and 23 samples of adjacent tissues of carcinoma were collected from patients with colorectal carcinoma, who did not receive radiotherapy or chemotherapy before surgery. Among the colorectal carcinoma samples, 21 were well-differentiated, 22 intermediate-differentiated, and 17 poor-differentiated. Of the 60 patients, 36 had serosal infiltration and 21 had lymph node metastasis. The expression of p73 mRNA in colorectal carcinoma was detected by in situ hybridization, and that of p73 protein was determined using immunohistochemistry. Quantification analysis was carried out by using the SmartScape software. The results were expressed as the mean gray level, which is negatively correlated with the level of expression. Results The expression of p73 mRNA and protein in the colorectal carcinoma were significantly higher than those in the adjacent tissues (mean gray levels: 0.549 ± 0.080 and 0.481 ± 0.091 in the carcinoma tissues vs. 0. 703 ± 0.081 and 0.701 ± 0.068 in the adjacent tissues). The mean gray level of p73 mRNA and protein were 0.463 ± 0.082 and 0.472 ± 0.099 in the poor- and intermediate-differentiated carcinoma, 0.549 ± 0.088 and 0.535 ± 0.058 in the well-differentiated carcinoma; 0.517 ± 0.106 and 0.528 ± 0.100 in the cases with serosal infiltration, 0.602 ± 0.079 and 0.560 ± 0.058 in the cases without serosal infiltration; 0.513 ± 0.083 and 0.462 ± 0.105 in the cases with lymph nodes metastasis; and 0.607 ± 0.083 and 0.595 ± 0.080 in the non-metastatic cases. The differences between the groups were statistically significant (P < 0.05). Conclusions The expression of p73 increases in colorectal carcinoma samples, and correlated with the biological behavior of the cancer. It could be a prognostic indicator for colorectal carcinoma.
