摘    要:目的 Plackett-burman试验(P-B试验)方法联用正交试验优选蛹虫草多糖最佳提取工艺.方法 在单因素试验的基础上,通过P-B试验筛选出影响蛹虫草多糖提取率的显著因素,通过3因素3水平正交试验,获得蛹虫草多糖的最佳提取工艺.结果 通过P-B试验得出料液比、提取时间及提取温度是影响蛹虫草多糖提取率的显著因素,正交试验结果显示蛹虫草多糖最佳提取工艺为:料液比1∶62.5、提取时间3 h、提取温度90 ℃、提取次数为2次、乙醇浓度95%、沉淀时间24 h,提取率为2.145%.结论 P-B试验通过比较因子水平的差异与整体的差异来确定因子的显著性,减少了试验次数.

Abstract:Objective To optimize the experimental conditions of extracting polysaccharides in cordyceps militaris.Methods Taking extraction yield as the index to optimize the production process,and combining the orthogonal design method with Plackett-Burma based on the single factor experiment.Results The significant factors influencing the cordyceps extraction yield was material to liquid ratio,extracting time and extractingtemperature.The optimal production technology as follow:the ratio of cordyceps militaris and water was 1∶62.5,and extracting timewas 3 h,extractingtemperaturewas 90 ℃,extracting times was 2 times,ethanol concentration 95%,settling time for 24 hours,the extraction yield of 2.0767%.Conclusion Plackett-burman test could determine the significance of factorby comparing the difference among the factorslevels and the overall,and reduce the testnumber.
