宫内生长迟缓大鼠成年期发生胰岛素抵抗的机制

摘    要:目的:探讨宫内生长迟缓(intrauterine growth retardation,IUGR)的雄性大鼠肝胰岛素受体(insulin recep-tor,IR)、胰岛素受体底物-1(insulin receptor substrate-1,IRS-1)、胰岛素受体底物-2(insulin receptor substrate-2,IRS-2)、磷脂酰肌醇3激酶(phosphatidyl inositol 3-kinase,P13K)等胰岛素信号传导蛋白及胰岛素样生长因子-1(insulin-like growth factor-1,IGF-1)的表达及与其发生胰岛素抵抗的关系.方法:用雌性大鼠妊娠期半定量饥饿法建它IUGR大鼠模型.RT-PCR法检测出生后12周龄雄性大鼠肝IR,IRS-1,IRS-2,PDK和IGF-1 mRNA的表达,免疫组织化学法检测肝IR,IQRS-1和IRS-2蛋白的表达.结果:12周龄IUGR雄性大鼠肝IR mRNA表达(0.41±0.06)较对照组(0.62±0.11)下降(P<0.05),而蛋白的表达(6.21±0.57)较对照组(6.69±0.47)无明显降低(P>0.05);IRS-1 mRNA(0.77±0.20)较对照组(1.32±0.42),IRS-2 mRNA(1.05±0.280)较对照组(1.48±0.40),IRS-1蛋白(2.15±0.23)较对照组(5.96±0.38),IRS-2蛋白(4.33±0.29)较对照组(7.08±0.35)的表达均受到抑制(P<0.05);PDK mRNA(1.12±0.19)的表达与对照组(1.18±0.24)差异无统计学意义(P>0.05);肝IGF-1mRNA的表达(0.55±0.12)较对照组(1.22±0.34)明显降低(P<0.05);相关分析显示IGF-1 mRNA在肝的表达降低与IRS-1 mRNA(r=0.821,P<0.05)和IRS-2 mRNA(r=0.643,P<0.05)的表达减少有关.结论:IUGR雄性大鼠成年期肝胰岛素受体信号传导蛋白IRS-1和IRS-2的表达受剑明显抑制,并导致肝IGF-1表达降低,可能与其生后的牛长迟缓及胰岛素抵抗有关;肝IR和PDK的表达与IUGR大鼠胰岛素抵抗的发生无明显关系.
