尼可地尔-乙基纤维素固体分散物的药物释放动力学的研究

摘    要: 目的:研究乙基纤维素固体分散物颗粒及片剂的释放动力学及释放机制。方法:测定尼可地尔一乙基纤维素固体分散物颗粒及片剂的释放曲线,并用3种模型方程——零级动力学方程、一级动力学方程、Higuchi模型方程进行线性拟合,根据所得方程的相关系数的大小顺序,确定颗粒及片剂的释放动力学及释放机制。结果:颗粒释放动力学拟合顺序为:零级动力学模型、Higuchi模型、一级动力学模型;片剂释放动力学拟合结果的优劣顺序为:Higuchi模型、一级动力学模型、零级动力学模型结论:颗粒释放遵从零级动力学模型方程,为表面扩散机制;片剂释放遵从Higuchi方程,为骨架扩散机制。

Abstract:OBJECTIVETo study the release kinetics and releas e mechanisms ofethylcellulose solid dispersion granules and tablets. METHODSThe dissolution curves of nicrandil-ethylcellulose soliddispersion granules and tablets were measured. The release data were fitt ed with threemodel equations. The release kinetics and release mechanisms of g ranules and tablets werejudged by the sequence of correlation coefficients of t hree fitting equatuions. RESULTSThe sequence of fitness(from good to bad) of granul es dissolution model was zero-order model, Higuchi model,first-order model. The sequence of fitness (from good to bad) of tablets disslution modelwas Hig uchi model, first-order model, zero-order model. CONCLUSIONThe drug release ofgranules was surface diffusio n mechanism, following zero-order release kinetics. The drugrelease of tablet s was matrix diffusion mechanism, following Highchi square rootequation.
