IL-17调控DLBCL小鼠VEGF表达对肿瘤生长的作用

目的:建立弥漫大B细胞淋巴瘤（DLBCL）小鼠模型并接种体外培养的Th17细胞，对比接种组与对照组IL-17及VEGF表达水平的变化，分析IL-17水平与VEGF表达的相关性及其与肿瘤发生发展的关系。方法:Mini MACS免疫磁珠分离系统分离纯化BALB/c小鼠脾来源的CD4+CD62L+ nave T细胞，加入anti-CD3、anti-CD28、TGF-β、IL-6进行Th17细胞体外培养，SUDHL-4细胞进行传代培养后接种于SCID小鼠建立DLBCL小鼠模型，对荷瘤小鼠进行 Th17 细胞接种，ELISA法检测DLBCL小鼠肿瘤组织中IL-17及血管内皮生长因子（VEGF）的表达。结果:接种Th17细胞后DLBCL小鼠IL-17表达水平较对照组升高，VEGF表达水平较对照组降低；随着肿瘤病程进展IL-17表达水平降低，VEGF表达水平升高。IL-17与VEGF之间存在显著负相关。结论:IL-17可下调VEGF的表达从而影响DLBCL的疾病进程，联合检测IL-17和VEGF表达水平对判断DLBCL疾病状态及进程更有参考价值。

IL -17 regulates the expression of VEGF in DLBCL mice and its influence on tumor growth

Objective:Established DLBCL mice model and received Th17 cells adoptive immunity,compare the expression level of IL-17 and VEGF in the Th17 cells immunity group and the compare group,analyse the relation between IL-17 and VEGF expression and the relation with tumor growth.Methods:The CD4+CD62L+ T cells purified by MACS were stimulated under cytokine conditions including anti-CD3,anti-CD28， TGF-β and IL-6 in vitro,transfer and culture SUDHL-4 cell strain and then inoculate the SCID mice,establish DLBCL mice models,inject Th17 cells to mice to get the adoptive immunity.ELISA tested IL-17 and VEGF expression in the tumor tissue.Results:The expression level in Th17 cells immunity group was higher than in compare group,but the expression of VEGF in Th17 group was lower than in compare group.According to the progress of the DLBCL,IL-17 expression level decreased and VEGF expression level increased.There was significant negative correlation between IL-17 and VEGF.Conclusion:IL-17 can down-regulate VEGF expression,expression of IL-17 and VEGF have reference value to judge the disease situation and the progress of DLBCL.
