RP-HPLC同时测定丙戊酸钠和卡马西平的血药浓度

目的：建立同时测定血清中卡马西平（CBZ）及丙戊酸钠（VPA）的高效液相色谱法。方法：0.1mL样本血清经1mL二氯甲烷：异丙醇（95∶5）混合液提取,以氟西泮（FZP）为内标,流动相为乙腈：磷酸盐缓冲液（30∶70）,色谱柱Agilent ZORBAX Extend-C18（4.6mm×150mm,5μm）,检测波长210nm。结果：在一定浓度范围内（CBZ：0.47～30.00μg.mL-1,VPA：1.25～80.00μg.mL^-1）被测药物与内标的峰面积之比与浓度呈良好的线性关系,平均回收率分别为98.5%、97.5%,日内和日间精密度均小于3%。结论：本方法操作简便,精密度好,回收率高。经临床用于常用抗癫痫药血药浓度的监测,其结果稳定可靠,可为临床进行群体药动学研究及实施个体化给药提供真实可信的资料。

Simultaneous Determination of Carbamazepine and Sodium Valproate in Serum by RP-HPL

OBJECTIVE：To develop a RP-HPLC method for simultaneous determination of carbamazepine （CBZ） and sodium valproate（VPA） in human serum. METHODS： 0.1 mL sample serum was extracted with 1 mL dichloromethane∶ isopropanal （95∶5） with Flurazepam （FZP） served as an internal standard. The mobile phase was composed of acetonitrile-phosphate buffer （30∶70）. The Agilent ZORBAX Extend-C18 （4.6 mm×150 mm,5 μm） column was used and the wavelength was set at 210 nm. RESULTS： A good linear relation was noted between the peak area ratio of the test drug to internal standard and the concentration at a certain range （CBZ： 0.47～30.00 μg·mL^ -1 ;VPA： 1.25～80.00 μg·mL^ -1 ）. The average recoveries were 98.5% and 97.5%,respectively;and the intraday RSD and inter-day RSD were all less than 3%. CONCLUSION： The method is simple in operation with good precision and higher recovery,and the results was proved to be stable and reliable in determination of antiepileptic drugs,which provided real and reliable data for the study of population pharmacokinetics and the implementation of individual dosage regimen.
