海昆肾喜对慢性肾衰竭大鼠肾脏保护作用及其机制探讨

摘    要:目的:观察海昆肾喜对慢性肾衰竭模型大鼠的肾脏保护作用,并初步探讨其机制。方法:将Wistar雄性大鼠随机分为3组,即:(1)正常对照组(n=15);(2)CRF模型组(n=15),给予腺嘌呤200 mg.kg-1.d-1;(3)海昆治疗组(n=15),给予腺嘌呤200 mg.kg-1.d-1同时给予海昆200 mg.kg-1.d-1,持续喂养4周,于每周末称重、收集大鼠尿标本,测定24 h尿蛋白;第4周末取血,仪器法检测血肌酐、尿素氮,并留取肾组织,切片HE染色,免疫组化方法半定量分析肾组织中结缔组织生长因子(CTGF)的表达。结果:海昆治疗组大鼠各时间段24 h尿蛋白均明显低于模型组(P〈0.05),血肌酐、尿素氮水平明显低于模型组(P〈0.05)。海昆治疗组大鼠肾组织CTGF蛋白表达明显低于模型组(P〈0.05)。结论:应用海昆肾喜治疗可以减少腺嘌呤致肾衰竭大鼠的尿蛋白排泄,降低血肌酐、尿素氮,减轻肾纤维化程度,其肾脏保护作用机制可能与抑制CTGF蛋白表达有关。

Abstract:OBJECTIVE:To observe the renoprotective action of Hai Kun Shen Xi(HKSX) in chronic renal failure model rats and explore the action mechanism.METHODS: Male Wistar rats were randomly divided into three groups: normal group(n = 15),CRF model group(adenine 200 mg.kg-1.d-1)(n = 15),and HKSX group(adenine 200 mg.kg-1.d-1 + HKSX 200 mg.kg-1.d-1)(n = 15).The rats were fed with drugs for 4 consecutive weeks.At each weekend,the rats were weighed and rate urine samples were collected for detection of 24-hour urine volume.Blood samples were collected at the 4th weekend for determination of serum creatinine and urea nitrogen(UN),and the renal tissue sample was collected and section of renal tissues was HE-stained,with the expression of connective tissue growth factor(CTGF) in the nephridial tissue detected by immunohistochemistry.RESULTS: In CRF + HKSX group,the levels of 24-h urinary protein,serum creatinine and UN were all significantly lower than in the CRF model group(P 0.05).Furthermore,the expression of CTGF in nephridial tissue was significantly lower in CRF + HKSX group than in the CRF group(P 0.05).CONCLUSION: HKSX can reduce urine protein excretion and down-regulate serum creatinine and UN levels,ameliorate renal fibrosis in rats with adenine-induced chronic renal failure.Its renoprotective action is probably achieved through inhibition on protein expression of CTGF.
