灰树花多糖对人外周血单个核细胞白细胞介素-2及干扰素-γ表达的影响

摘    要:目的探讨灰树花多糖(GRN)对人外周血单个核细胞(PBMCs)白细胞介素-2(IL-2)、干扰素-γ(IFN-γ)表达的影响。方法使用不同浓度GRN与人PBMCs共培养,对照组以不加GRN的PBMCs培养相同时间。RT-PCR检测人PBMCs中IL-2 mRNA的表达,应用流式细胞术检测Th1细胞中IFN-γ的表达。结果经GRN处理的人PBMCs中,IL-2 mRNA的表达较对照组呈显著增高(P〈0.01);Th1细胞中IFN-γ的表达与对照组相比也显著增高(P〈0.01)。结论 GRN可提高PBMCs中IL-2 mRNA及IFN-γ的表达,提高细胞免疫功能。

Abstract:Objective To investigate the effects of griflolan on immunological function of normal human PBMCs.Methods PBMCs treated with different concentration GRN,culture for 24 hours,control group of PBMCs cultured the same time without GRN;use RT-PCR to detect the level of IL-2 mRNA in PBMCs,the levels of interferon-gamma of CD4+ T lymphocytes cells in PBMCs were measured by folw ctrometry.Results The levels of IL-2 mRNA and INF-γ in PBMCs that treated with GRN were significantly higher than those in the group control(P0.01).Conclusion Grifolan might increase the level of IL-2 and INF-γ of normal human PBMCs,promote the cells immune function of human.
