来氟米特活性代谢产物对肿瘤坏死因子-α诱导肾小管上皮细胞内核因子-κB活性的影响

摘    要:目的:探讨来氟米特活性代谢产物对TNF-α诱导肾小管上皮细胞内NF-κB活性表达的影响。方法:培养的肾小管上皮细胞为空白组;对照组为肾小管上皮细胞加TNF-α5000U/ml共同培养;干预组先于上皮细胞中加不同浓度来氟米特的活性代谢产物(A771726)培养不同时限,30min后再加入TNF-α5000U/ml。EMSA法检测各组NF-κB活性表达的变化。结果:A771726在不同浓度及时限均可抑制TNF-α诱导的肾小管上皮细胞内NF-κB活性表达,且呈剂量与时限依赖性。结论:来氟米特活性代谢产物可抑制TNF-α诱导的肾小管上皮细胞内NF-κB的活性表达。

Effect of Leflunomide on NF-κB Activity Expression in TNF-α Stimulating Proximal Tubular Cells
Abstract:Objective: To explore whether or not the function of leflunomide in decreasing NF-κB activity expression in overload TNF-α stimulating proximal tubular cells. Method: Cultured cells without stimulation as placebo; cultured cells were incurbed with TNF-α 5 000 Uml as the control; cultured cells were incurbed with different concentration of leflunomide for different hours as the interfere; EMSA was used to observe NF-κB activity expression. Results: Active leflunomide metabolite A771726 can decrease NF-κB activity expression. The decreasing was time and dose dependent. Conclusion: Leflunomide can decrease NF-κB activity expression in overload TNF-α stimulating proximal tubular cells.
