对赤芍醇提浸膏和水提液治疗胆汁淤积型肝损伤的药效筛选

目的:筛选有治疗胆汁淤积型肝损伤作用的赤芍提取物。方法:将小鼠随机分成正常对照组、模型组、赤芍醇提浸膏高、中、低剂量组,水提液高、中、低剂量组和阳性药赤丹退黄颗粒组;用α-萘异硫氰酸酯建立胆汁淤积型小鼠模型,连续给药6天后,检测血清ALT、BIL、ALP、TBA等指标,并进行病理组织观察和半定量分析。结果:赤芍醇提浸膏能显著降低血清DBIL、ALP、ALT和AST值(P<0.05或P<0.01),并能明显改善α-萘异硫氰酸酯引起的组织损伤(P<0.01),中剂量作用与阳性对照药相似;而水提液没有治疗作用(P>0.05)。结论:赤芍治疗胆汁淤积型肝炎的有效药物成分主要在赤芍醇提浸膏中。

Pharmacodynamic screening Radix Paeoniae Rubra extract for treatment of cholestatic hepatic injury

Objective: To screen the effective extract of Radix Paeoniae Rubra on cholestatic hepatic injury.Methods: All mice were randomly divided into nine groups : the control group,the model group,the extracta sicca groups of high dose,medium dose and low dose,the water extract groups of high dose,medium dose and low dose,and the Chidan granula group.The cholestatic hepatic injury animal models were made by alpha-naphthylisothiocyanate(ANIT).All animals were given the drugs or normal saline for six days.The serum indexes of ALT,BIL,ALP,TBA were tested and the pathological changes of the livers were analyzed by semiquantitative method.Results: The extracta sicca decreased the levels of DBIL,ALP,ALT and AST in serum(P<0.05 or P<0.01),and it also significantly improved the liver injury induced by ANIT(P<0.01).The extracta sicca group of medium dose had the same effect as the Chidan granula group.There was no effect in the water extract groups(P>0.05).Conclusion: The pharmacodynamic ingredients healing cholestatic hepatic injury are in the extracta sicca of Radix Paeoniae Rubra.
