甲亢肝阳上亢证大鼠肾上腺组织蛋白质组的双向电泳分析

摘    要:目的观察甲亢肝阳上亢证大鼠与正常大鼠肾上腺中蛋白质的差异表达,以期发现诊断甲亢肝阳上亢证的标志物,为进一步探讨甲亢肝阳上亢证的分子机制提供依据。方法左旋甲状腺素(L-T4)和附子汤灌胃造成甲亢肝阳上亢证模型,应用蛋白质组学技术比较模型组与正常组中肾上腺的差异蛋白质,结合生物信息学对其进行分析鉴定。结果模型组与正常组相比有33个蛋白质上升和6个蛋白质下降,进一步质谱鉴定,其中6个蛋白质为GrpE蛋白同系物1、60kDa热休克蛋白、脂酰辅酶A脱氢酶、钙网织蛋白前体、醛脱氢酶、异戊酰辅酶A脱氢酶。结论甲亢肝阳上亢证大鼠肾上腺中具有特异表达的蛋白质,经质谱鉴定后发现,其蛋白质的变化可能与甲亢肝阳上亢证的形成密切相关。

Abstract:Objective To further explore the molecular mechanism of the hyperthyroidism syndrome of hyperactivity of liver-YANG and to find the diagnostic biomarker by observing differences of protein expressions in adrenal gland in the hyperthyroidism rats with hyperactivity of liver-YANG and normal rats.Methods Animal models were established by ip levo thyroxine(L-T4) and guizi tang.Both the quantitative and qualitative changes of the protein expressions were compared between the control group and treated group by protein techniques.Results 33 proteins increased and 6 proteins decreased their expressions in model group compared with treated group,some of which included GrpE protein homolog 1,60 kDa heat shock protein,calreticulin precursor,Acyl-Coenzyme A dehydrogenase,Isovaleryl-CoA dehydrogenase and Aldehyde dehydrogenase.Conclusion These proteins that changed their expression levels may be involved in the development of the hyperthyroidism with hyperactivity of liver-YANG.
