摘    要:目的:研究不同浓度三氧化二砷(As2O3)对人骨髓增生异常综合征难治性贫血伴原始细胞过多型(MDS-RAEB)细胞株MUTZ-1细胞增殖的影响,并探讨其可能的作用机制。方法:采用MTT法分析细胞生长曲线,用细胞形态学观察细胞凋亡。结果:低浓度As2O3(0.05~0.25μmol/L)对MUTZ-1细胞无明显生长抑制作用,高浓度As2O3(1.0~20.0μmol/L)对MUTZ-1细胞有明显生长抑制作用,且呈浓度、时间效应-依赖关系(P<0.01)。结论:低浓度As2O3(0.05~0.25μmol/L)对MUTZ-1细胞无明显生长抑制作用。高浓度As2O3(1.0~20.0μmol/L)对MUTZ-1细胞有明显生长抑制作用,且呈浓度、时间效应依赖关系。

Abstract:Objective To investigate the inhibition effect of As2O3 on the growth of human MDS-RAEB cell line MUTZ-1 cells and to explore the possible cellular and molecular mechanisms.Method The experiment use the method of MTT to investigate the inhibition effect of arsenic trioxide(As2O3)on the growth of human MDS-RAEB cellline MUTZ-1 cells and to explore the possible cellular and molecular mechanism.Results Low concentration of As2O3(0.05~0.25μmol/L)had no marked growth inhibition effect on MUTZ-1 cells,high concentration of As2O3(1.0~20.0μmol/L)had marked growth inhibition effect on MUTZ-1 cells.After 48h treatment with2.0μmol/L As2O3.Conclusion Low concentration of As2O3(0.05~0.25μmol/L)had no marked growth inhibition effect on MUTZ-1 cells,high concentration of As2O3(1.0~20.0μmol/L)had marked growth inhibition effect on MUTZ-1 cells.
