黑逍遥散对AD小鼠脑组织AchE,ChAT,MAO活性的影响

摘    要:目的研究黑逍遥散对氢溴酸东莨菪碱所致老年性痴呆(AD)模型小鼠脑组织AchE,ChAT,MAO活性的影响。方法将60只SPF级小鼠随机分为5组,即空白对照组、模型对照组、黑逍遥散组、逍遥散组及精简方组,每组12只。空白对照组、模型对照组给予生理盐水,黑逍遥散组、逍遥散组及精简方组分别给予相应生药水煎液,连续灌胃45 d,其中给药第41~45 d,除空白对照组外,其余各组小鼠均腹腔注射氢溴酸东莨菪碱2.0 mg/(kg.d)进行造模。第45 d给药及造模后处死小鼠,取其脑组织,测定小鼠脑组织匀浆中AchE,ChAT,MAO活性。结果黑逍遥散、逍遥散及精简方均可显著升高ChAT活性,降低AchE及MAO活性(P0.01或P0.05),但以黑逍遥散作用最为明显。结论黑逍遥散能较好地预防治疗AD,其作用可能与调节中枢胆碱神经递质及单胺类神经递质的活性有关。

Abstract:Objective To research the effects of Heixiaoyao San on the activity of AchE, ChAT and MAO in brain tissue of model mice with AD induced by scopolamine hydrobromide. Methods 60 mice were randomly divided into blank control group, model control group, Heixiaoyao San group, Xiaoyao San group and Jingjian Fang group ,every group had 12 mice. The mice of blank control group and model control group were given normal saline, the mice of other groups were given relevant herbs decoction. All mice had been administrated for 45 days. In the forty first to forty fifth day, except blank control group, the mice of other groups were injected scopolamine hydrobromide of 2.0 mg/( kg · d) in abdominal cavity to make the model of AD. After giving medicines and making model in forty fifth day, killed all mice and taken brain tissue, determined the activity of AchE, ChAT and MAO in brain tissue of mice. Results Heixiaoyao San, Xiaoyao San and Jingjian Fang could significantly increase the activity of CHAT, and decrease the activity of AchE and MAO( P 〈 0.01 or P 〈 0.05 ), but the effect of Heixiaoyao San was strongest. Conclusion Heixiaoyao San can prevent and treat AD, the effect could relate to adjusting the activity of central cholinergic nerve transmitter and monoamine nerve transmitter.
