摘    要:目的 观察β-七叶皂苷钠对胶质瘤细胞增殖及血管内皮生长因子(VEGF)表达的影响.方法 将10、20、30、40、50 mg/L的β-七叶皂苷钠作用于体外培养的U251胶质瘤细胞并设立空白对照组,噻唑蓝(MTT)比色法检测细胞增殖,流式细胞仪分析细胞周期的分布;免疫组织化学检测增殖细胞核抗原(PCNA)和VEGF表达.结果 与对照组比较,β-七叶皂苷钠明显抑制U251细胞的增殖和生长,其作用呈时间效应关系和剂量效应关系(P<0.05).β-七叶皂苷钠作用后13251细胞PCNA、VEGF表达降低,呈时间依赖性(P<0.05).结论 β-七叶皂苷钠可抑制胶质瘤细胞增殖和VEGF表达,对胶质瘤的瘤周水肿可能具有治疗作用.

Abstract:Objective To study the effect of sodium aescinate on vascular endothelial growth factor (VEGF) expression and cell proliferation in gliomas. Methods In vitro cultured human U251 glioma cells were incubated with 10,20, 30,40 and 50 mg/L sodium aescinate. Methyl thiazolyl tetrazolium (MTr) assay was used to evaluate cell growth. Cell cycle distribution was evaluated by flow cytometry. The expression of proliferating cell nuclear antigen (PCNA) and VEGF proteins was detected by immunohistochemistry. Results Compared with the control group, sodium aescinate significantly inhibited the growth and proliferation of U251 cells in a time- and dose-dependent manner (P < 0.05). After treatment with sodium aescinate, the expression of PCNA and VEGF was inhibited significantly in a time-dependent manner (P <0.05). Conclusion Sodium aescinate maybe have an therapitical effect on glinma growth and paratumor edema through down-regulating the VEGF expression and cell proliferation.
