肝肾阴虚型甲亢和脾肾阳虚型甲减患者血清蛋白质组的比较分析

摘    要:[目的]通过对肝肾阴虚型甲亢和脾肾阳虚型甲减患者血清蛋白质组的分析,阐述肝肾阴虚和脾肾阳虚的物质基础。[方法]将正常人、甲亢和甲减患者血清用亲和层析法除去其中的高丰度蛋白质,再用双向电泳技术分析不同组样品蛋白质组的变化,所获的差异蛋白质用质谱进行鉴定。[结果]与正常人相比,肝肾阴虚型甲亢患者血清中RBP和TTR的表达量降低,而IGF-2、FDA-C、NF-κB及PTP表达量升高;脾肾阳虚型甲减患者TTR表达量升高,NHE-1、ZAG、UCH、IGF-2和MHC–II蛋白则表达量下降。肝肾阴虚型甲亢和脾肾阳虚型甲减相比,ProapoA-Ⅰ和TTR表达量降低,而NDU、SCDH/SCR、PK、Cyt P450蛋白、IGF-2和IRF-5表达增加。[结论]肝肾阴虚型甲亢患者机体代谢活跃,而脾肾阳虚型甲减患者则机体代谢减低,合成代谢的速度分解代谢的速度。肝肾阴虚型甲亢血清IGF-2表达升高,脾肾阳虚型甲减则正好相反,说明血清IGF-2具有成为中医辨证肝肾阴虚和脾肾阳虚血清标志物的可能性。

Analysing the Serum Proteomics Between Hyperthyroidism of Yin deficiency in Liver and Kidney and Hypothyroidism of Yang Deficiency in Spleen and Kidney

Abstract:[Objective] Material basis of yin deficiency of liver and kidney and yang deficiency of spleen and kidney were studied by analysing the difference of serum proteome between the patients of hyperthyroidism and hypothyroidism.[Methods] The high abumdance proteins in the serum of normal people,hyperthyroidism patients and hypothyroidism patients were removed with Proteo-Extract Albumin/IgG Removal Kit and the proteins were separated by two-dimensional electrophoresis and anysised with software.The different proteins were identified with mass spectrum.[Results] Compared with the normal people,the expression of retinol binding protein and transthyretin decreased while the expression of insulin growth factor2,aldolase C,NF-κB,protein-tyrosine phosphatase increased in the serum of yin deficiency of liver and kidney patients,and the expression of Na/H hydrogen exchanger isoform1,Zn-alpha-2-glyoprotein,ubiquitin C-terminal hydrolase,insulin growth factor-2,MHC-Ⅱdecreased while the expression of transthyretin increased in the serum of yang deficiency of spleen and kidney.When compared with the hypothyroidism patients of yang deficiency of spleen and kidney,the expression of proapolipoproteinApoA-I and transthyretin decreased,and the expression of NADH ubiquinone dehydrogenase,short-chain dehydrogenase/reductase,pyruvate kinase,cytochrome P450,interferon regulatory factor 5 and insulin growth factor 2 increased in yin deficiency of liver and kidney hyperthyroidism.[Conclution] The energy metabolism in the patients of yin deficiency of liver and kidney was active.In contrast,there were low energy metabolism in the patients of yang deficiency of pleen-kidney.The expression of insulin growth factor 2 has contrary variance in this experiment,so it might be a serum marker for differentiating zheng in traditional chinese medicine.
