大蒜素对血管紧张素Ⅱ诱导血管平滑肌细胞增殖的抑制作用

目的 观察大蒜素对血管紧张素Ⅱ诱导血管平滑肌细胞增殖的抑制作用。方法 采用培养的兔血管平滑肌细胞，应用^3H～TdR掺入法，观察在血管紧张素Ⅱ促进VSMC增殖过程中，大蒜素对血管平滑肌细胞DNA合成的影响及其时间效应。结果 血管紧张素Ⅱ可促进处于静止状态的兔血管平滑肌细胞DNA的合成，36h时细胞DNA的合成达到高峰。大蒜素对血管紧张素Ⅱ诱导的血管平滑肌细胞增殖有明显的抑制作用，并呈现出明显的浓度依赖关系，随着大蒜素表度的升高其对血管平滑肌细胞增殖的抑制作用也逐渐增加，在24h时，10．00g／L的大蒜素对血管平滑肌细胞增殖的抑制率为13．46％。结论 大蒜素可抑制血管紧张素Ⅱ诱导兔血管平滑肌细胞的增殖，并存在一定的量效依赖关系及时间反应性。

Inhibitory effects of garlic extract on proliferation of cultured vascular smooth muscle cells induced by angiotension Ⅱ

[Objective] To investigate the inhibitory effects of garlic extract on proliferation of cultured vascular smooth muscle cell induced by angiotension II(Ang II). [Methods] The rabbit vascular smooth muscle cells were cultured and the inhibitory effect of garlic extract on DNA synthesis in vascular smooth muscle cells stimulated by Ang II was assayed at different time and different concentration by using 3H-TdR incorporation in vitro. [Results] Ang II could significantly promote DNA synthesis of quiescent rabbit vascular smooth muscle cells with a maximal response time at 36 h. Garlic extract could significantly inhibit DNA synthesis of vascular smooth muscle cells in a dose-dependent manner, with a maximal response at a concentration of 10.00 g/L and at 24 h that the inhibitory rate was 13.46%. [Conclusion] Garlic extract could significantly inhibit DNA synthesis of ribbit vascular smooth muscle cells induced by Ang II and in a dose and time dependent manner.
