聚集素、P-糖蛋白在原发性肝细胞癌中的表达及其相关性研究

目的探讨聚集素（CLU）在原发性肝细胞癌（HCC）中的表达及其与P-糖蛋白（P—gP）表达的相关性。方法采用免疫组化法检测凋亡抑制基因CLU在HCC（41例）、肝硬化（10例）和正常肝脏（10例）组织中的表达及P—gp在HCC中的表达。结果CLU在41例HCC组织中阳性表达率为82．93％（34／41），10例肝硬化和10例正常肝脏组织中表达阴性或弱阳性。在HCC组织中，CLU阳性表达与年龄、性别及有无合并肝硬化无相关性（P〉0．05），而与Edmondson分级明显相关（P〈0．05）。P—gP在HCC组织中的阳性表达率为70．73％（29／41）。在HCC组织中，CLU的表达与P—gP表达呈正相关（P〈0．05）。结论CLU在HCC中呈高表达，与临床耐药密切相关，有望成为HCC靶向治疗的一个新靶点。

The expression of clusterin and P-glycoprotein in hepatic cell carcinoma and its relationship

Objective To study the expression of clusterin （CLU） and P-glycoprotein （P-gp） in hepatic cell carcinoma （HCC） and its relationship. Methods The expression of CLU gene in the tissue of H CC （41 cases）, cirrhosis（ 10 cases） and normal liver （ 10 cases） and the expression of P-gp in the tissue of HCC were detected by immunohistochemical method. Results The positive expression rate of CLU was 82.93% （34/41） in HCC, and negative or weak positive were detected in cirrhosis and normal liver tissue. The positive expression rate of CLU was significantly correlated with Edmondson＇s histological grade （P 〈 0.05 ）, but had no correlation with patients＇age, sex and cirrhosis（P 〉 0.05 ）. The positive expression rate of P-gp in HCC was 70.73%（29/41 ）.There was significant correlation between the expression of CLU and P-gp （P 〈 0.05）. Conclusions The over expression of CLU is significantly associated with clinical muhidrug resistance in HCC. It should be a potential target for treatment of HCC.
