摘    要:目的观察硫酸软骨素蛋白多糖Aggrecan在大鼠脊髓半横断损伤(HSCI)后的表达变化。方法 20只成年SD大鼠随机分为假手术对照组和HSCI 3、7和14 d组,建立成年大鼠HSCI(胸9~10)模型;取损伤位点头尾段胸9、胸10节段制作石蜡切片,用免疫组织化学ABC法和图像分析技术检测脊髓内Aggrecan的表达。结果 Aggrecan阳性产物主要分布于细胞外基质中,神经元和神经胶质细胞中也有分布;损伤后脊髓内Aggrecan表达较对照组明显增加。结论 HSCI后Aggrecan表达明显增加,可能参与损伤后轴突生长被抑制这一过程。

Abstract:Objective Changes of Aggrecan of expression in rats spinal cord following hemisection of spinal cord injury have been studied to provide the experimental evidence for the mechanisms of regeneration and repair after spinal cord injury.Methods 20 adult healthy Sprague-Dawley rats were divided into sham-operated control group and 3days,7days,14days spinal cord injury groups at random.The spinal cords were hemisected between T9 and T10.The T9~10 spinal cord was isolated and sectioned.Aggrecan was observed by immunohistochemical staining.And the immuno-positive gray scale of immunoreactant was detected with computer image analysis technique.Results Aggrecan positive products were mainly distributed in neurons and neuroglial cells in spinal cord and ECM for the control group.The immuno-positive expression for Aggrecan in three spinal cord injury groups was all increased as compared with cortrol group.Conclusion The expression of Aggrecan increases after HSCI,it may mutually play inhibitory effect on axon regeneration.
