3,6-二羟基黄酮对多种肿瘤细胞的增殖抑制及凋亡诱导作用

摘    要:目的 观察3,6-二羟基黄酮对乳腺癌细胞株MCF-7(雌激素受体阳性ER+)、MDA-MB-231(雌激素受体阴性ER-)、直肠癌细胞株LoVo、DLD-1和前列腺癌细胞株PC3增殖和凋亡的影响.方法 MTT法检测3,6-二羟基黄酮对不同肿瘤细胞生长存活的影响,计算半数抑制浓度(IC50);DAPI染色观察3,6-二羟基黄酮对肿瘤细胞生长状态及形态的影响,Annexin V/PI双染流式细胞仪检测细胞凋亡变化.结果 增殖检测结果表明,3,6-二羟基黄酮对于MCF-7、MDA-MB-231、LoVo、DLD-1和PC3细胞增殖均表现出显著抑制作用,48 h IC50值分别为16.1、15.7、29.8、12.8和17.8μmol/L;细胞形态观察和凋亡检测结果表明,10 μmol/L的3,6-二羟基黄酮作用24 h,5种肿瘤细胞的生长状态和形态均发生显著变化,大量细胞表现出凋亡或死亡细胞的形态特征,细胞凋亡率均显著升高.结论 3,6-二羟基黄酮具有显著的抗肿瘤效应,对于多种肿瘤细胞具有很强的增殖抑制和凋亡诱导作用.

Abstract:Objective To observe the effects of 3,6-dihydroxyflavone on the proliferation and apoptosis in breast cancer cell lines MCF-7 (ER+) and MDA-MB-231 (ER-),colorectal carcinoma cell lines LoVo and DLD-1,prostatic cancer cell line PC3.
