长链非编码RNASPRY4-IT1在脑胶质瘤中的表达及对U87细胞功能的影响

摘    要:目的探讨长链非编码RNA SPRY4-IT1在脑胶质瘤中的表达,以及对胶质瘤细胞系U87细胞生物学功能的影响。方法收集我科40例胶质瘤手术标本(包括肿瘤组织和瘤周组织),荧光实时定量PCR(qRT-PCR)检测40对标本中SPRY4-IT1的表达情况;用慢病毒介导shRNA干扰SPRY4-IT1表达后,分别采用CCK-8法、流式细胞仪和Transwell法检测其对细胞增殖、凋亡和迁移的影响。结果 SPRY4-IT1在肿瘤组织中的表达量为11.73±0.55,高于瘤周组织的9.14±0.59,差异有统计学意义(P0.05)。SPRY4-IT1表达被干扰后,胶质瘤细胞株U87迁移能力明显降低,差异有统计学意义(P0.05),但细胞增殖无明显变化,差异无统计学意义(P0.05),细胞凋亡变化亦不明显,差异无统计学意义(P0.05)。结论 SPRY4-IT1在胶质瘤组织中高表达,并且能促进U87细胞迁移,可能成为胶质瘤诊断和判断预后的重要分子标记物。

Abstract:Objective To investigate the SPRY4-IT1 expression levels in glioma and the role of SPRY4-IT1 in glioma U87 cell biological functions. Methods Quantitative real-time polymerase chain reactions for SPRY4-IT1 expression were performed in 40 pairs of glioma tissues and adjacent tissues in our department. Lentiviral-mediated SPRY4-IT1 knoekdownin U87 was further to investigate its role in glioma cell biological functions. Cell growth, apoptosis and migration were tested in vitro by CCK-8 assays, flow eytometry and transwell assays, respectively. Results SPRY4- IT1 expression was significantly overexpressed in cancer tissues (P 〈 0.05 ). Knockdown of SPRY4- ITI expression reduced cell migration(P 〈0.05) , but have no significant changes in cell growth(P 〉 0.05 ) and cell apoptosis ( P 〉 0.05 ). Conclusions SPRY4-IT1 was overexpressed in glioma tissues and promoted the migration of U87 cells. The expression of long non-coding RNA SPRY4-IT1 may play an important role in the process of glioma development and may be developed as a useful biomarker for the diagnosis and prognosis of glioma.
