辛二酰苯胺异羟肟酸联合顺铂对裸鼠肾上腺皮质癌的实验研究

目的：通过荷瘤裸鼠模型研究辛二酰苯胺异羟肟酸（SAHA）联合顺铂对肾上腺皮质癌的疗效及毒副作用。方法：建立肾上腺皮质癌裸鼠荷瘤模型，随机分为对照组、顺铂组、SAHA组和SAHA联合顺铂组（联合组）4组，连续21天经腹腔注射药物后处死裸鼠，测定瘤体直径、瘤重，计算移植瘤的体积及肿瘤生长抑制率，同时检测裸鼠的红细胞、白细胞、血小板、血清总胆固醇、谷丙转氨酶、尿素氮、肌酐含量，对数据进行统计分析。结果：与对照组相比，各治疗组均显示出肿瘤体积缩小及肿瘤生长抑制率增加（P〈0．05）；治疗组间两两相比，联合组显示出最明显的肿瘤体积缩小及肿瘤生长抑制率增加（P〈0．05）。顺铂组、联合组较对照组均出现白细胞计数下降，血清谷丙转氨酶、尿素氮及肌酐水平升高（P〈0．05）；而在顺铂组与联合组的比较中，上述四项指标差异均无统计学意义（P〉0．05）；SAHA组与对照组相比较，血清谷丙转氨酶水平升高，但白细胞及尿素氮、肌酐水平差异均无统计学意义（P〉0．05）。结论：在荷瘤裸鼠模型实验中，sAHA可增强顺铂对肾上腺皮质癌疗效而毒副作用无明显增加。

Effects of SAHA combined with cisplatin on adrenocortical carcinoma in nude mice model

Objective： To study the effects of suberoylanilide hydroxamie acid （SAHA） combined with cispla- tin on adrenocortical carcinoma in nude mice model. Method： SW-13 adrenocortieal carcinoma nude mice model was constructed by hypodermic injection of SW-13 cell suspension. All the nude mice were randomly divided into four groups： control group, cisplatin group, SAHA group and cisplatin plus SAHA group. The counting of red blood cells, white blood cells and platelets, serum levels of cholesterin, glutamic pyruvic transaminase, urea nitro- gen and creatinine, volume and wet weight of tumor were recorded. Then the tumor growth inhibition rate and tumor volume were analyzed by SPSS 13.0. Result： There were statistically significant differences in the tumor growth inhibition rate and tumor volume between the control group and the other three treatment groups （ P 〈 0.05）, especially in cisplatin plus SAHA group （ P 〈0.05）. The decrease of white blood cell counting and the in- crease of serum levels of glutamic pyruvic transaminase, urea nitrogen and creatinine were found in the groups used cisplatin （ P 〈0.05）, but there were no statistical differences between cisplatin group and cisplatin plus SA HA group （ P 〉0.05）. The increase of serum levels of glutamic pyruvic transaminase was observed in the group SAHA, but there was no statistical difference in white blood cell counting, serum levels of urea nitrogen and cre- atinine （ P 〉0.05）. Conclusion： In nude mice model with adrenocortical carcinoma, SAHA combined with cispla tin can lead to synergistic effects but little side effects.
