| 摘 要： | 目的探讨食管鳞癌中凋亡相关蛋白激酶1（DAPK1）基因CpG岛甲基化及其蛋白表达与食管癌的相关性。方法收集食管鳞癌组织58例及癌旁上皮组织30例。采用免疫组织化学链霉素抗生物素蛋白过氧化物酶（SP）法检测DAPK1蛋白表达，甲基化特异性聚合酶联反应（MSP）法检测基因CpG岛甲基化状态。结果DAPK1在癌组织的阳性表达率明显低于癌旁组织（36．2％ vs 68．3％，P〈0．05）；癌组织中DAPK1甲基化率明显高于癌旁组织（60．3％VS33．3％，P〈0．05），提示DAPK1基因启动子甲基化与DAPK1蛋白表达呈负相关（X^2=18．362，P=0．000，r=-0．563）。结论DAPK1基因启动子甲基化减少其蛋白的表达，可能与食管癌的发生发展相关。 |
