还原型谷胱甘肽在不同潮气量机械通气对大鼠肺细胞凋亡的影响

摘    要:目的探讨还原型谷胱甘肽对不同潮气量机械通气大鼠肺细胞凋亡的影响。方法将30只健康雄性SD大鼠随机分成对照组(A组,n=6)、正常通气组(B组,n=6)、过度通气组(C组,n=6)、正常通气组+还原型谷胱甘肽(D组,n=6)及过度通气组+还原型谷胱甘肽(E组,n=6)五组,D、E组在机械通气前5min阴茎背静脉注射还原型谷胱甘肽,各组大鼠通气时间均为4h。凋亡细胞的检测使用末端脱氧核苷酸转移酶介导的dUTP缺口标记技术,用免疫组织化学技术检测肺组织核转录因子-κB(NF—KB)的表达。ELISA法检测肺匀浆中肿瘤坏死因子-α(TNF-α)变化。结果C组肺组织中TNF-α含量与D、E比较明显增加,C组肺组织NF-κB的表达组与D、E组比较明显增加。C组肺组织中细胞凋亡明显高于D、E组。结论还原型谷胱甘肽能减少机械通气损伤中引起的肺组织的细胞凋亡。

Abstract:Objective To observe the effects of reduced glutathione on pulmonary apoptosis indueed by different tidal volume mechanical ventilation in rats. Methods Thirty SD rats were randomly divided into five groups: control group (group A, n = 6), normal ventilation group (group B, n = 6), hyperventilation group ( group C, n = 6), normal ventilation + reduced glutathione group ( group D, n = 6 ), hyperventilation + reduced glutathione group ( group E, n = 6). The Nuclear factor-κB ( NF-κB) expression in pulmonary endothelium was assessed by histochemical staining;the methods of ELISA was used to determined the level of tumor necrosis factor alpha ( TNF -α). Apoptosis was investigated by TdT - mediated dUTP flick end labeling. Results The expression of the NF -κB, TNF - α was significantly higher in group C than in group A and group B, group D, group E. In group C pulmonary apoptosis were obviously than in group D and E ( P 〈0.01 ). Conclusions Reduced glutathione can decrease pulmonary apoptosis ventilation -induced lung injury.
