乙酰肝素酶和nm23蛋白在恶性黑素瘤中的表达及意义

摘    要:目的探讨恶性黑素瘤的发生和侵袭转移机制,检测乙酰肝素酶、nm23在恶性黑素瘤中的表达及意义。方法应用免疫组织化学sP法检测30例恶性黑素瘤、30例黑素细胞痣和20例正常皮肤组织中乙酰肝素酶和nm23蛋白的表达。结果乙酰肝素酶在恶性黑素瘤组织中表达的阳性率(63.33%)明显高于其在黑素细胞痣中的表达(6.67%),且其在正常皮肤组织中仅在皮脂腺腺体细胞中表达,其余细胞中不表达,两者比较差异有统计学意义(P〈0.05);nm23在恶性黑素瘤组织中表达的阳性率为36.67%,明显低于其在黑素细胞痣中的表达(86.67%),且在正常皮肤组织中为90.00%,两者比较差异有统计学意义(P〈0.05)。结论乙酰肝素酶和nm23与恶性黑素瘤的发生有关,提示恶性黑素瘤组织中乙酰肝素酶和nm23的检测可作为预测肿瘤的指标之一。

Abstract:Objective To clarify the molecular mechanism of development,invasion and metastasis of human malignant melanoma, we detect the expression of heparanase and rim23 in malignant melano- ma. Methods The expression of heparanase and nm23 in 30 malignant melanoma, 30 melanocytic nevus and 20 normal skin were detected by SP immunohistochemistry. Results Positive rates of heparanase in malignant melanoma (63.33%) were significantly higher than that in normal skin. Positive rates of nm 23 in malignant melanoma (36.67%) were significantly lower than that in normal skin (90.00%) and melanocytic nevus(86.67% ). Both of the expression of heparanase and nm23 in metastasize malignant mela- noma were greatly different to the no metastasis ones. Conclusions Heparanase and nm23 are closely related to the metastasis of malignant melanoma, and may be the predictors of the metastasis.
