摘    要:目的 体外培养足细胞,探讨高糖刺激下足细胞凋亡与smad通路的关系,并研究来氟米特是否可以通过该通路来抑制足细胞凋亡.方法 传代培养人肾小球足细胞后分组实验,用western blotting法检测smad2/3蛋白及p-smad2/3蛋白的表达,并用流式细胞仪检测足细胞的凋亡.结果 高糖刺激下足细胞表达p-smad2/3蛋白及足细胞凋亡均增加;加入抑制剂及来氟米特后,二者均显著减少.其中来氟米特组较高糖组足细胞凋亡率下降了(24.0±2.3)%,P<0.05.结论 来氟米特能部分阻断高糖环境下smad通路所致的足细胞凋亡.

Abstract:Objective Cultured podocytes in vitro, explore the relationship of podocytes apoptosis and smad pathway stimulated by high glucose and to examine whether leflunomide can inhibit the foot of the pathway to apoptosis. Methods Nestin training human glomerular podocyte before divided into experimental groups, with the western blotting assay smad 2/3 protein and p-smad 2/3 protein expression and by flow cytometry at different time points podocyte apoptosis. Results Podocytes high glucose stimulated p-smad 2/3 protein increased, and podocyte apoptosis increased. Compared with the high glucose group, after adding inhibitors and leflunomide, podocytes p-smad 2/3 proteins were reduced,and podocyte apoptosis were also reduced. Conclusions Leflunomide partially blocked high glucoseinduced podocyte smad pathway of apoptosis could reduce the degree of renal sclerosis.
