基质金属蛋白酶-2、9及其抑制剂-1在大肠腺癌中表达的研究

目的:探讨人大肠腺癌组织中MMP-2、MMP-9和TIMP-1的表达及其临床意义。方法:采用免疫组织化学方法(SP)检测MMP-2、MMP-9和TIMP-1在30例大肠癌组织及相应切缘正常组织中的蛋白表达。结果:大肠癌组织中MMP-2、MMP-9及TIMP-1阳性表达明显高于正常切缘组织(P<0.05),并与肿瘤分化程度、浸润深度、Dukes分期以及有无淋巴结转移密切相关(P<0.05);MMP-9和TIMP-1表达明显相关(r=0.4924,P<0.05)。结论:MMP2、MMP9、TIMP1的过度表达与大肠腺癌的侵袭、转移有密切关系,其联合检测有助于评估大肠腺癌恶性生物学行为及预后。

Expression of MMP-2, MMP-9 and TIMP-1 in human colorectal cancer

Objective:To study the significance of matrix metalloproteinase-2(MMP-2), matrix metalloproteinase-9 (MMP-9) and their inhibitor tissue inhibitor of metalloproteinase TIMP-1 expressions in human colorectal cancer. Methods:The expressions of MMP2,MMP9 and TIMP1 in 30 primary colorectal tumors and adjacent noncancerous tissues were detected by immunohistochemical S-P method. Results:The positive rates of MMP-2,MMP-9 and TIMP-1 in colorectal cancer were significantly higher than those in adjacent noncancerous tissues(P<0.05),and which were closely associated with tumor differentiation ,Dukes stages, progression of invasion depth and lymph node metastasis (P<0.05).A close correlation was present between MMP-9 and TIMP-1 expression(r=0.4924,P<0.05). Conclusion: Over expressions of MMP2,MMP9,TIMP-1 are obviously correlated with both invasion and metastasis in colorectal cancer , Combined detecting of which may be helpful to determining colorectal cancer biological behavior and prognosis.
