凝血酶对大鼠脑出血血肿周围组织的毒性损伤研究

摘    要:目的 :研究脑出血血液凝固过程中产生凝血酶的神经毒性作用。方法 :90只大鼠分成 :对照组、脑出血组、脑出血水蛭素干预组、凝血酶组和凝血酶 +水蛭素干预组。术后 48h观察大鼠神经功能缺损评分 (NDS) ,检测血肿周边组织髓过氧化物酶 (MPO)的活性、脑水肿含量及TUNEL阳性细胞数量。结果 :局部注入重组水蛭素显著减轻了脑出血大鼠神经功能缺损 ,降低局部MPO活性、减少脑水分含量及TUNEL阳性细胞数量 ;脑内直接注入凝血酶后局部脑水分含量升高 ,MPO活性升高 ,并可检测到大量TUNEL阳性细胞。结论 :凝血酶与脑出血后的脑水肿、白细胞浸润和神经细胞DNA损伤有关

Toxic Effect of Thrombin to Prihematomal Tissue Following Intracerebral Hemorrhage
Abstract:Aim:To investigate the toxic effect of thrombin on perihematomal tissue injuries in intracerebral hemorrhage (ICH).Methods:Ninety rats were divided into five groups:ICH groups,ICH plus hirudin groups,thrombin groups,thrombin plus hirdin groups,and sham operation groups.48 h later,all rats were decapitated after neurological deficit scores of rats were evaluated,then brain water content and MPO activities were measured and the expression of TUNEL positive cells in perhimatoma were detected.Results:Compared with ICH group,TNEL positive cells,brain water content,and MPO activities in perihematoma were all decreased significantly when recombinant hirudin was injected immediately into hematoma (P<0.01),so did the NDS of rats.When 10 U thrombin was injected into the right caudate nucleus,there were higher content of brain water and MPO activities in local tissue,a number of TUNEL positive cells were detected as well.Conclusion:Thrombin following ICH may be involved in preihematomal edema,leukocuyte infiltration and neuronal DNA damage.
