摘    要:目的 评估重组大鼠胰岛素样生长因子-1(rrIGF-1)聚(乳酸-羟基乙酸)共聚物(PLGA)缓释微球对2型糖尿病GK大鼠种植体周围骨形成的影响.方法 将20只雄性GK大鼠进行糖尿病造模成功后随机分为2组,分别给予加载包裹rrIGF-1的PLGA缓释微球(治疗组,n=10)和加载包裹与rrIGF-1等量安慰剂的PLGA缓释微球(糖尿病组,n=10)处理,再以10只未做处理的SD大鼠作为对照组.在所有大鼠胫骨内植人种植体,治疗组和糖尿病组大鼠同时分别加载等量相应种类的PLGA缓释微球.术后4、5、8周在种植体周围局部采血,采用酶联免疫吸附法(ELISA)检测血清骨钙素(OCN)、骨特异性碱性磷酸酶(B-ALP)及Ⅰ型前胶原羧基前肽(PICP)含量.结果 种植体植入术后4周,糖尿病组和治疗组的血清OCN、B-ALP和PICP水平均明显低于对照组(P均<0.05);术后5周,糖尿病组的OCN和B-ALP水平明显低于对照组和治疗组(P均<0.05),糖尿病组和治疗组的PICP水平也均明显低于对照组(P<0.05),治疗组的OCN水平明显高于术后4周(P<0.05),对照组的PICP水平则明显低于术后4周(P<0.05);术后8周,糖尿病组大鼠的OCN和B-ALP水平明显低于对照组和治疗组(P均<0.05),糖尿病组和治疗组大鼠的血清PICP水平均明显低于对照组(P均<0.05).结论 rrIGF-1 PLGA缓释微球局部加载可促进GK大鼠种植体周围骨形成.
