窄吸收窗药物缓释制剂的研发进展

摘    要:综述窄吸收窗药物缓释制剂开发相关的最新进展和策略。药物(或制剂)与其吸收部位的接触时间是由药物通过胃肠道转运的速度所决定的。而缓释制剂要达到有效的缓释效果,必须要持续地将药物释放到吸收部位。目前用于提高窄吸收窗药物缓释制剂生物利用度的策略主要是减少药物在非吸收窗部位的破坏、拓宽药物的吸收窗和延长药物在吸收窗的滞留时间。

Abstract:The development and R&D strategies of sustained release preparations for narrow absorption window drugs were reviewed. An effective sustained release preparation has to release drug to absorptive sites persistently and the time that the drug is in contact with its preferred absorptive site is determined by the transit of it through the gastrointestinal tract. Absorption windows limit the bioavailability of the sustained release preparations. The methods designed to improve the bioavailability of the controlled release preparations for narrow absorption window drugs fall into three different categories: 1.preventing the destruction of drugs at non-absorptive sites, 2.widening the absorption windows and 3.prolonging the time that the drugs are at the absorption window.
